| In order to screen out the acute renal injury biomarkers of dogs with specificity and high sensitivity,two models of acute renal injury were established in dogs with nephrotoxicity and ischemia-reperfusion in this article.And concentration changes of four kinds of renal injury markers were detected.The correlation between the changes of four kinds of renal injury markers and the degree of renal injury was verified by histopathology.The changes of serum creatinine and urea nitrogen were used to verify whether the four kinds of renal injury markers could be used as early diagnostic indexes of kidney.The results showed that the concentrations of RBP,NAG,KIM-1 and NGAL in the urine and blood were significantly increased with the prolongation of the time of injection,which had a good correlation with the degree of renal injury.The concentration of RBP in the urine was significantly higher on the 4th day after injection of gentamicin sulfate,and the RBP in the blood was significantly increased on the fifth day,which was earlier than that of creatinine and BUN,could be used as early diagnosis of acute renal injury indicators.The concentration of NAG in urine and blood was significantly higher than that of CREA and BUN before the injection of gentamicin on the third day,which could be used as early diagnosis index of canine kidney injury.The concentration of KIM-1 in the urine began to increase significantly on the first day of injection of gentamicin sulfate,and the concentration of KIM-1 in the blood began to increase significantly on the third day after gentamicin sulfate injection,which was slightly earlier than CREA and BUN and could be used as early diagnosis of renal injury indicators.The concentration of NGAL in urine was significantly higher than that in gentamicin sulfate on day 2,and the concentration of NGAL in blood was significantly increased on the fifth day after the injection of gentamicin sulfate,which could be used as a sensitive marker for the early diagnosis of canine nephrotoxicity renal injury.The increasing time of NGAL concentration in blood was earlier than that of CREA and BUN,and their sensitivity was quite.The results of the experiment in the model of renal ischemia-reperfusion injury,showed that 4 kinds of the biomarkers increased in the blood and urine,and their concentration was correlated positively with the time of renal artery occlusion prolong.And their increasing time was significantly earlier than the CREAand BUN.So these four kinds of renal injury biomarkers could be used as acute renal injury early diagnosis of sensitive indicators by renal ischemia and reperfusion.To explore the therapeutic effect of intermittent hemodialysis on acute renal failure in dogs,this study established the acute renal failure model induced by gentamicin sulfate and the acute renal failure model induced by ethylene glycol poisoning at first.After the clinical examination and laboratory results proved the replication of the models were successful,the effect of intermittent hemodialysis therapy was verified by treating the the models by intermittent hemodialysis therapy.The results showed that intermittent hemodialysis therapy could quickly remove the accumulation of creatinine and urea nitrogen in the blood,and not only quickly correct the electrolyte and acid-base balance disorder,but also cause no loss of protein,which could help dogs remove toxic and harmful substances from the kidney further poison and provide enough time for the recovery of kidney units.The results proved that intermittent hemodialysis could be promoted in clinical use. |
