| Feijoa sellowiana Berg also known as Feijiguo,Feiyouguo,is a myrtle family,a genus evergreen shrub or a small tree.It is a new type of fruit tree that is edible,medicinal and ornamental.feijoa can adapt to the climate of Mianyang,Sichuan Province,and maintain its fruit quality and ornamental characteristics.It is a tree species with great development potential.At present,there is no systematic feijoa-harvesting seedling breeding technology and base in China.In order to promote the rapid development of the feijoa fruit industry and realize large-scale promotion and large-scale cultivation as soon as possible,it is particularly important and urgent to study the seedling breeding technology of feijoa.In this paper,tissue culture was carried out on the 1-year-old young stern tip of three varieties of feijoa,and total phenolic and total flavonoids content in the stem tip of each treatment were analyzed.To explore the relationship between total phenolics and total flavonoids content and the browning of the feijoa tissue culture.Initial conclusions are drawn as follows:(1)Through the different disinfection time treatment of explants,the pollution rate,browning rate and callus rate of 7d and 30 d were observed and counted.Analyze and compare the differences of different disinfection time on the tissue culture of stem tip of three varieties.The results showed that the Opall Star pollution rate and browning rate were the lowest,the pollution rate and browning rate were the lowest in 6min disinfection time,and the callus induction rate was the highest.(2)Through the comparison of different disinfection time,the optimal disinfection time was 0.1% mercury disinfection for 6min,and different concentrations of plant growth regulators were added to the MS medium for callus induction.The results showed that Coolidge had the lowest pollution rate and browning rate,and Opal Star had the highest call rate.Among the four mediums,No.1 medium MS+6-BA 1.0 mg/L had the lowest contamination rate;No.4 medium MS+6-BA 2.0 mg/L+NAA 1.0 mg/L had the lowest browning rate No.3 culture Base MS+6-BA 2.0 mg/L has the highestcall rate.(3)The 1 year old shoot tips were used as explants,and they were sterilized under sterile conditions in 4 callus induction medium supplemented with antioxidant Vc.The control was not added with any antioxidant.After 30 days of cultivation,the pollution rate,browning rate and callus rate were counted.The results showed that the addition of antioxidant Vc had the most obvious inhibitory effect on the browning of MS+6-BA 1.0 mg/L in No.1medium and MS+6-BA 1.0 mg/L+NAA 1.0 mg/L in No.2 medium.The browning rate of No.1 medium MS+6-BA 1.0 mg/L was the largest.The inhibition effect of Coolidge browning is most obvious.(4)The disinfection time had a significant effect on the total phenolics and total flavonoids content in the stern tip of feijoa tissue culture,and the content gradients of total phenols and total flavonoids were similar.In general,the total phenolic and total flavonoids content was the highest in the 3min disinfection time,and the total phenolic and total flavonoids content was the lowest in the 9min disinfection time.The 9min disinfection time inhibited the production of total phenols and total flavonoids content.(5)had a significant effect on the total phenolics and total flavonoids content in the stern tip of feijoa tissue culture.The total phenol and total flavonoids in the No.4 medium were the highest,and the total phenolics and total flavonoids in the No.3 medium were the lowest.Coolidge was in 3 The content of total phenol and total flavonoids was the highest in the medium,and the content of total phenol and total flavonoids in the medium No.2 was the lowest.The content of total phenol and total flavonoids in the medium No.3 was the highest,and the content of total phenol and total flavonoids in the medium No.4 was the lowest.In general,MS+6-BA 2.0 mg/L had the highest content of total phenols and total flavonoids,and No.2 medium MS+6-BA 1.0mg/L+NAA 1.0 mg/L had the lowest content of total phenolics and total flavonoids.(6)The effect of adding Vc on the content of total phenol and total flavonoids in the stern tip offeijoa tissue culture was significant.The No.2medium MS+6-BA 1.0 mg/L+NAA 1.0 mg/L had the greatest influence,and the No.2 medium had lower total phenol and total flavonoids in Unique and Coolidge.The total phenol and total flavonoids of Opal Star The content isincreased.The content of total phenol and total flavonoids in other media decreased to a certain extent,among which Unique decreased the most,but Coolidge showed a decreasing trend in the four mediums,and the trend was the most stable among the three varieties.(7)The factors affecting tissue culture browning are very complicated.Based on the regression analysis of the browning rate of the stern tip of three varieties of Unique,Opal Star and Coolidge,and the total phenolic and total flavonoids content of the tissue cultured stern tip,There was a significant positive correlation between total phenolic content and browning rate(0.01<P<0.05),and there was a significant positive correlation between total flavonoid content and browning rate(P<0.01),indicating that the content of phenolic substances was on the stem of the fruit.The browning of culture has a significant effect.As the browning rate of tissue culture increases,the content of total phenols and total flavonoids also increases. |
