| Cytospora chrysosperma is important phytopathogens in Xinjiang,causing canker disease on Poplar,Salix,Apple tree,Ulmus pumila and other hosts.C.chrysosperma which collected from different hosts showed a certain difference on cultural characteristics.In order to clarify the diversity,phylogenetic relationship and genetic difference of C.chrysosperma of Xinjiang.The cultural characteristics,Biolog-FF and ISSR marker were used to analyze the genetic diversity of C.chrysosperma which were collected on 5 different plants and 6 different regions.The conclusions in the current study are below.(1)47 C.chrysosperma strains were collected on 5 different plants and 6 different regions.Based on the cultural characteristics,47 strains were divided into 15 different types.(2)Biolog-FF system with 95 kinds of carbon source was used to exploit the metabolism feature of C.chrysosperma.The results showed that 882 strain used the most carbon sources and the activity of it was rel-atively high,the lowest is 812-1.The carbon source type of carbohydrate,carboxylic acid and itsderivant,p-olymer and amino acid were utilized by C.chrysosperma.There had some carbon source only used by 6 str-ains,1074-2(L-alaninamide,L-asaragine),891-1(L-lactic acid,L-phenylalanine),847(Bromosuccinic aci-d),896(L-fucose),934(Adenosine),740(P-hydroxyphenylacetic acid,Succinamic acid and L-asparagine).(3)Cluster analysis of 47 C.chrysosperma strains which based on genetic similarity coefficient showed that 47 strains of C.chrysosperma were divided into 2 clusters,the first cluster included 14 different type strains with different colony morphology,the second cluster are belong to the type of white with small dense fruiting bodies.Based on the genetic distance,the strain was clustered,the relationship of Tacheng,Hami,Tulufan,Wujiaqu,Changji were near.(4)By comprehensive analyzing the cultural characteristics,physiological-biochemical characteristic and ISSR marker,the results showed that the strains which used carbon source independently had different phenotype,740(White with fruiting bodies in group),1074-2(Grey without fruiting bodies),891-1(White without fruiting bodies and fluffy texture),847(Dark grey without fruiting bodies),896(Dark grey with small dense fruiting),934(Orange without fruiting bodies).812-1,904,739,626,919-1,933 strains using the fewest carbon sources and had lowest activity,they were belong to the type of white with dense fruiting bodies. |
