Endophytic Antagonistic Bacillus is a kind of microorganism that is established in healthy plants and has a harmonious relationship with plants.It has many biological functions,such as disease resistance,growth promotion,and plant stress tolerance.In recent years,the classification status,bacteriostasis activity and growth promoting effect of many endophytic bacteria have been studied.In this study,antifungal endophytic bacteria were isolated from tomato,pepper and other plants,and the types,antibacterial activity and antibacterial characteristics of the antifungal substances secreted by them were studied.The results were as follows:1.Endophytic bacteria against pathogenic fungi were isolated from different parts of tomato and pepper plants,and screening.The results showed that the 19 strains of endophytic bacteria have better antifungal activity between 50.3%-86.7%,on Alternaria alternate.And the inhibitory rate of Fq23,Fxj82 XJ5 were 86.7%,80%and75%,respectively.The inhibitory rate of X8,X18,X25 and Fq153 strains against Alternaria alternate was 63.3%.2.using the method of organic solvent extraction,silica gel column chromatography and detected the growth rate of Fxj5 and Fxj82 strains of antibacterial substances were crude and purified.The results showed that the Fxj5 were chromatography and obtained four fractions,and the inhibition rates were 29%,58%,20%,28%,respectively.The Fxj82 were obtained four fractions too.For the inhibition rates were 54%,30%,20%,20%,respectively.Indicated that the antibacterial substances had been got better separation and laid the foundation for further separation and purification3.Antifungal proteins which produced by XJ5,XJ6,FQ23 and X8 strains were the main antifungal substance.To research the antifungal protein,salted the antimicrobial substances by 40%,50%,60%,70%,80%saturation of ammonium sulfate.And then,the antifungal activity of proteins was measured through growth rate method,alternaria alternata was the indicator bacteria.Screened optimal ammonium sulfate concentration of salting,and compared the antibacterial activity of four kinds of antagonistic proteins.The results showed that the best ammonium sulfate fractionation concentrations of XJ5 and XJ6 and FQ23 and X8 were 80%,40%,60%,80%,respectively.Among the four kinds of Bacillus sp.,antifungal proteins produced by X8 had good antibacterial activity.The Inhibitory against Alternaria alternata and Alternaria solani and Fusarium oocysporum were 71.68%,83.46%,87.94%,respectively.4.Alternaria alternata,for instructions bacteria were determined to four kinds of antimicrobial protein pathogenic bacteria and the influence of hyphae spore germination.The results of the study show that:XJ5 antifungal protein processing after 48 h reach maximum,OD260,OD280 value are 1.748 and 1.829,XJ6 antimicrobial protein processing after 24 h reach maximum,OD260,OD280 value were 1.654,1.8,X8 antimicrobial protein processing after 48 h reach maximum,OD260,OD280 value were 2.087,1.993,FQ23 antimicrobial protein processing after 48 h reach maximum,OD260,OD280 value were 1.933,1.79,the results show that extraction of bacillus antimicrobial protein on the cell membrane permeability caused by bacteria influence,and cause bacteria in the body of nucleic acids and proteins coming out,research also shows that the Alternaria alternata by the antifungal protein processed become deformity,the bacteria conidium cannot normally germinate.
