Natural rubber industry is a very important economic industry all over the world.Hainan province is the major plant area in China.One of the rubber tree diseases which is regard as severely endanger the natural rubber’s yield worldwide is rubber tree powdery mildew.Rubber tree powdery mildew spread by Oidium heveae B.A.Steinmn from the air.Oidium heveae B.A.Steinmn severely endanger rubber trees’ burgeen,inflorescence and leaf blade.Seriously,it can cause leaf blade off early,delayed rubber tapping,all of these seriously affect rubber trees’ growth and yield.In the early stage of rubber tree powdery mildew,it can be hardly seen with the naked eye,it will be out of control once it breaks out on a full scale,early forecasting technique is most significant to rubber tree powdery mildew’s prevention.Our research team has established relatively mature laboratory pathogenic bacteria’s detecting techniques on rubber tree powdery mildew,i,e.LAMP detecting techniques.In terms of the advantages of LAMP such as fast speed and visibility,this thesis preliminarily quests its application potential in rubber tree powdery mildew’s early forecast,and provides scientific basis for effectively prevent and control rubber tree powdery mildew in production.This study carries out preliminary improvement and optimization on DNA extraction method of Oidium heveae,with the aid of glycerol by changing the osmotic pressure inside and outside the cell walls of powdery mildew spores to the wall effect,simply and roughly extract rubber tree powdery mildew’s DNA.The expanded results of PCR showed that the simplified DNA extraction method established in this study could extract trace amounts of the spore DNA of powdery mildew.And it can be detected by LAMP technology.Based on this DNA extraction method,different amounts of spore DNA were extracted from rubber trees,and measure the DNA concentration.A series of group data were obtained,and via statistical analysis,a regression prediction mathematical model was established,y = 17.49x+ 62.78 and R squared = 0.884.On this basis,in the laboratory where room temperature is 25℃ Quantitative inoculation of Oidium heveae in young tissue of rubber trees,and continue observation,record the start time(symptomatic appearance),status,obtained a series of group data,statistical analysis of data obtained,A regression mathematical model was established for predicting the amount of spores and incidence of palbicans,y =11.091n(x)+46.06,R squared = 0.861.Simulate forest disease process indoor,set the temperature for the rubber tree powdery mildew of the susceptible temperature is 25 and 80%humidity,capture spore powder with a portable the spore powder capyurer,extract its DNA,detect the DNA extracted through established LAMP technology,utilize established regression prediction model to predict later incidence,realize early predict for rubber tree powdery mildew.Provides scientific basis for realizing field’s fast and exact predict,early and accurate predictions are great meaningful for effective prevention and control of rubber tree powdery mildew and save prevention and control costs.
