| Sophora flavescens is a leguminous plant,root can be used as a medicine,and the main medicinal active ingredients are matrine and oxymatrine.Exogenous substances can regulate its growth,development and secondary metabolism.In this study,four exogenous substances(Gibberellin,Salicylic acid,Lysine,Brassinolide)were selected for seed soaking and seedling leaf spraying of Sophora flavescens;By analyzing germination rate,seedling SPAD value,photosynthetic gas exchange parameters,biomass and total matrine content of Sophora flavescens,the effects of different exogenous substances on seed germination,photosynthetic physiology and total matrine accumulation of Sophora flavescens were studied.The research results are as follows:(1)Effects of exogenous substances on seed germination of Sophora flavescens.The gibberellin concentrations of 25,50 and 75 mg/L could significantly increased the germination potential and germination rate of Sophora flavescens seeds.The optimum gibberellin concentration for germination were 50mg/Lwere with the highest germination potential(46.0%)and germination rate(51.33%),which increased by 19.33% and 24.67%,respectively,compared to the control.Lysine and salicylic acid inhibited the seed germination of Sophora flavesne.0.1mg/L brassinolide promoted the germination of Sophora flavescens seeds,and the germination potential and germination rate increased by 13.76% and 18.75%,respectively,compared with the control.(2)Effects of exogenous substances on photosynthetic physiology of Sophora flavescens seedlings.After 20 d treated with 50mg/L gibberellin,the chlorophyll content and photosynthetic rate in the leaves of Sophora flavescens could be improved,and the chlorophyll content could be increased and the photosynthetic rate could be improved after the 4mmol/L lysine treated 10 d,and the promoting effect could still be shown after the cultivation of 41 d.After 20 days of 50 mg/L gibberellin,chlorophyll content and photosynthetic rate in Sophora flavescens leaves were increased;after 10 days of 4 mmol/L lysine culture,chlorophyll content and photosynthetic rate were increased,which could be promoted until 41 days of culture;after 20 and 41 days of 100 mg/L salicylic acid culture,chlorophyll content increased by 0.88% and 2.33%,but photosynthetic rate was lower than that of control.After the cultivation of Sophora flavescens with 0.1mg/L of brassinolide,the chlorophyll content in the leaves increased,which increased the photosynthetic rate.It indicated that the treatment of different exogenous substances could increase the chlorophyll content of leaves and enhance the photosynthetic efficiency of Sophora flavescens.(3)The effect of exogenous substances on the biomass of Sophora flavescens seedlings.Salicylic acid had no significant effect on the accumulation of root biomass in Sophora flavescens.of 2.0mmol/L~6.0mmol/L lysine,0.1mg/L brassinolactone and 25 mg/L gibberellin could significantly increased the biomass(dry weight)of Sophora flavescens roots,which increased by 18.27%-26.90%,21.75% and 7.02% respectively,compared to the control group,among which 4 mmol/L lysine had the best effect.(4)Effects of exogenous substances on total alkaloids of Sophora flavescens seedlings.The content of total alkaloids of Sophora flavescens varies among organswhich were the highest content in roots and the lowest content in stems.Salicylic acid and lysine increased the content of total alkaloids,4mmol/L lysine and 100mg/L salicylic acid have the most obvious effect,and the content of total alkaloids is 6.48 mg/g and 5.36mg/g,respectively,33.80% and 19.96% higher than the control.Gibberellin and brassinolide reduced the content of total alkaloids.(5)Seven alkaloid-related genes and two flavonoid-related genes were obtained by SSR analysis and gene excavation of transcriptome data of Sophora flavescens.leaves... |
