| In this experiment,the endogenous hormone content of tissue-cultured seedlings of Lyciumru ruthenicum Murr with different ploidy was determined by high performance liquid chromatography(HPLC),in order to find out the reasons for the difficulty of rooting polyploidy,and the morphological characteristics of Lyciumru ruthenicum Murr with different ploidy and the content of physiological active ingredients in plant leaves were also determined in order to predict the growth table of Lyciumru ruthenicum Murr with different ploidy in the field.The differences of related components in fruit were also discussed.The results show that:1.The contents of seven endogenous hormones including GA3,IAA,ABA,ZT,IBA,KT and 6-BA in leaves of tissue culture seedlings of Lyciumru ruthenicum Murr with different ploidy were determined by HPLC.There were significant differences in GA3,IAA,ABA,ZT,IBA and KT between diploid and polyploid Lyciumru ruthenicum Murr,among which the contents of ABA and KT increased with the increase of ploidy,while the contents of IAA,IBA and GA3 decreased with the increase of ploidy,while the contents of 6-BA in leaves of diploid and polyploid Lyciumru ruthenicum Murr had no significant difference.That is to say,the contents of ABA and KT in leaves of Lyciumru ruthenicum Murr L.are positively correlated with ploidy,and the contents of IAA,IBA and GA3 are negatively correlated with ploidy,while the contents of 6-BA are not correlated with ploidy.2.There were significant differences in plant height,stem diameter and leaf width between diploid and polyploid Lyciumru ruthenicum Murr in tissue culture,but there was no significant difference in leaf length.With the increase of ploidy,plant height became shorter,while stem diameter and leaf width increased significantly with the increase of ploidy.3.The content of chlorophyll,ascorbic acid and anthocyanin in Lyciumru ruthenicum Murr with different ploidy is different,the higher the ploidy is.the higher the content of anthocyanin is;the higher the ploidy is,the higher the content of anthocyanin is,the difference between octoploid and diploid is significant,while the content of tetraploid anthocyanin is not significantly different from Octoploid and diploid,but the higher the ploidy is,the higher the anthocyanin content is.The content of ascorbic acid in tetraploid plants was not significantly different from that in diploid and octoploid plants,but the higher ploidy was,the less ascorbic acid was.The content of ascorbic acid in octoploid plants was significantly different from that in diploid and octoploid plants.That is,there is a positive correlation between anthocyanin and chlorophyll content and ploidy,and a negative correlation between ascorbic acid content and ploidy.4.Due to the significant difference of endogenous hormone content,the suitable medium for the proliferation of Lyciumru ruthenicum Murr ploidy is different.The optimum medium for diploid was MS+0.15mg·L-1 ZT,in which the multiplication multiple of diploid was 6.65,while that of tetraploid and octoploid was only 3.75 and 3.65;in MS+1.0mg·L-1 GA3,the multiplication multiples of tetraploid and octoploid were 5.25 and 5.60,respectively,while that of control diploid was only 2.80.5.The rooting rate of tetraploid was the highest in MS+0.3mg·L-1 NAA medium,28.6%,while that of Octoploid was the highest in MS+0.5mg·L-1 NAA medium,40%. |
