| Deoxynivalenol(DON)is one of the most serious mycotoxin contamination in the diet,descend the feed intake of poultry,lower the immunity function,impact the health of intestine and create significant losses to poultry farming.There are many malpractices for traditional methods of detoxification.Adsorbent has no selectivity,and the weak polarity structure of deoxynivalenol molecular reduce adsorption efficiency of adsorbent.In this study,we supplemented lactic acid bacteria into diet which contaminated by deoxynivalenol and detected the effects of lactic acid bacteria for intestinal health of broilers,expect to find effective feasible solutions of detoxification.Exp.1 The effect of Lactic acid bacteria on performance and digestive absorbtion of broiler chickens challenged with deoxynivalenolA total of 192 one-day-old male chicks(AA)with similar body weight from a commercial hatchery were randomly divided into 4 groups consisting of 6 replicates with 8birds per replicate.Birds of each group received one of the following dietary treatments:1)control group(CON)fed basal diet;2)probiotic group(JM113)fed basal diet supplemented with freeze dried L.plantarum JM113 at 1×10~9 cfu per kg diet;3)Deoxynivalenol group(DON)fed basal diet contaminated with 10 mg DON per kg diet;4)Deoxynivalenol+probiotic group(DON+JM113)fed basal diet contaminated with 10 mg DON per kg diet and supplemented with freeze dried L.plantarum JM113 at 1×10~9 cfu per kg diet.The result showed that lactic acid bacteria has no significant effect on performance of broilers challenged with deoxynivalenol.Addition of L.plantarum JM113 resulted in higher villis,even in combination with DON compared with the DON group(P<0.05).DON decrease the relative expression of nutrient transporter gene r BAT in ileum and there is no significant difference among CON JM113 and JM113+DON.The present result show that lactic acid bacteria can effectively alleviate the adverse effects of DON on intestinal development,and improved the down-regulation of relative expression of the intestinal nutrient transporter of broiler caused by DON.Exp.2 The effect of Lactic acid bacteria on intestinal barrier function of broiler chickens challenged with deoxynivalenolAt d21 and d42 in the trial,the mucosa and segments of duodenum,jejunum and ileum were collected,the contents of cecum were collected for determination of microbial community structure and evolutionary changes by 16S rRNA at d42.Tight junction protein(ZO-1,Occludin and Claudin-1)and cytokines(IL-10,IL-12,TNF-αand INF-γ)were quantified by Real-time quantitative PCR and sIgA contents were detected by ELISA in mucosa.The result showed that DON down-regulate the relative expression of Claudin-1 in jejunum and there was no significant difference among CON,JM113 and JM113+DON.Lactic acid bacteria had no significant effect on the relative expression of Occludin and content of intestinal sIgA.DON down-regulate the relative expression of IL-12 compared with JM113 and JM113+DON and had no significant difference with CON.There is same dominant bacteria under phyla and family level.The present result show that lactic acid bacteria can effectively alleviate the adverse effects of DON on intestinal physical barrier and it can also regulate intestinal immune barrier. |
