The seeds of Rhizoma Polygonati have dormancy and are not easy to germinate.At present,there are some researches on the cause of dormancy of Dornata spermatorrhoids and the technology of breaking the seeds.The study found that the dormancy of the Rhizoma Polygonati seeds belongs to integrated dormancy,and the mechanical disturbance of the endosperm cells is one of the main causes of dormancy of the Rhizoma Polygonati seeds.In this paper,we studied the morphological characteristics of endosperm cells at different developmental stages,compared the morphology of endosperm cells in different producing areas,combined with changes in the activities of lignin metabolism-related enzymes and changes in the content of stone cells in different developmental stages and at different germination times.The underlying cause of seed dormancy.The main findings are as follows:(1)The study of the anatomical characteristics of sexual development of sexual organs of Rhizoma Polygonati revealed that the mechanical barriers of endosperm cells of Rhizoma Polygonati seed were formed.From late April to mid-May,the yellow flower buds emerge from budding to opening.At this time,the yellow and fine stamens gradually mature and mature.From late April to early June,the pistil of pterostilbene also develops orderly and completes the double fertilization in early June.June-August was the main period of development of each organ after fertilization.The fruit development of Rhizoma Polygonati was basically matured in the period of 8-10 months.During this period,a series of changes occurred in the Rhizoma Polygonati seed.Especially in the August-September period,the cell wall of the endosperm of the Rhizoma Polygonati was steepened and the cell contents disappeared.The formation of this specific structure makes the Rhizoma Polygonati seeds extremely rigid,resulting in poor permeability,affecting the normal water absorption and germination ofRhizoma Polygonati seeds.(2)Different environmental conditions have certain influence on the yellow spermatozoid endosperm cells.From the thickening degree of the Rhizoma Polygonati endosperm cell wall,the thick yellowish endosperm cell wall produced in Shaoguan of Guangdong Province,Lushan of Henan Province,and Huangshan of Anhui Province has a relatively shallow thickening,and each of them is not densely broken and wavy;Shaanxi Hanzhong,Jiangxi Xinfeng Rhizoma Polygonati thickening of the cell wall,the cell wall is more uniform,thicker parts more dense.(3)There was a certain correlation between the activity of lignin synthesis related enzymes and the content of stone cells and seed hardness in the Rhizoma Polygonati seed development.The activity curves of PAL,4CL,C4 H,and CAD were significantly different in Rhizoma Polygonati seeds.Among them,the activities of 4CL,C4 H,and PAL all increased with the maturity of seeds,indicating that these enzymes promoted lignin synthesis.The hard seed of Rhizoma Polygonati seeds played different degrees of promoting effect.During the seed development,the content of stone cells increased at first and then stabilized,and was positively correlated with the change of C4 H enzyme activity,and had a very significant positive correlation with the change of 4CL enzyme activity.It indicated that the seeds of Rhizoma Polygonati were accompanied by the synthesis of stone cells and lignin during the seed development.The correlation between the two was extremely high,which had significant impact on the seed hardness.(4)There was also a certain correlation between the activities of related enzyme of lignin synthesis and the content of stone cells and seed hardness in the germination of Rhizoma Polygonati.4CL activity decreased in the early stage of seed germination,PAL activity decreased in the late stage of seed germination,CAD and C4 H activity increased in the early stage of germination,and decreased in later stages.Overall,these four enzymes all responded to the germination of seeds.During the process of germination,there was a very significant positive correlation between the changes of stone cell content and the changes of CAD activity,which further demonstrated that the mechanical disorders of Rhizoma Polygonati seed were caused by stone cells and lignin.
