Effect Of Different Feeding Methods On The Deposition Of Fatty Acid Of Sunit Sheep And The Mechanism | | Posted on:2019-10-15 | Degree:Master | Type:Thesis | | Country:China | Candidate:L Yang | Full Text:PDF | | GTID:2393330566991144 | Subject:Food Science | | Abstract/Summary: | PDF Full Text Request | | The effect of different feeding patterns on the expression of fatty metabolism genes(SCD,FADS1,FADS2 and elove5),the contents of fatty acid and the ruminal microflora within different daipose tissue(intramuscular fat,tail fat and perirenal fat)and rumen fluid of 12 months Sunit Sheep were measured using Real Time polymerase chain reaction(RT-PCR),gas ? chromatography-mass ? spectrometry(GC-MS)and high-throughput sequencing.Moreover,the relationship among the abundance of fatty acids,the expression of metabolism genes and the ruminal microflora was explored.The results showed that abundance of C8:0 from the concentrated was significantly higher than the grazed in the different adipose tissue.The abundance of C10:0 from the grazed was significantly higher than the concentrated in intramuscular fat and tail fat.The abundance of C18:2 from the grazed was significantly higher than the concentrated in tail fat and perirenal fat.The abundance of C18:3 from the grazed was significantly higher than the concentrated in the different adipose tissue.The grazed was good for deposition of DHA and EPA,and the concentrated was good for deposition of saturated fatty acid in the different adipose tissue.The expression of SCD from the concentrated sheep were higher than the grazed in the different adipose tissues.The expression of FADS1 and FADS2 from the concentrated sheep were higher than the grazed in the intramuscular fat and perirenal fat.The expression of elove5 from the concentrated sheep were higher than the grazed in the tail fat and perirenal fat.The expression of FADS1 and FADS2 from the concentrated sheep were higher than the grazed in the intramuscular fat and perirenal fat.The gene expression of SCD and elove5 of the tail fat were higher than intramuscular fat and perirenal fat.The gene expression of FADS1 and FADS2 of the intramuscular fat were higher than tail fat and perirenal fat.As to the relationship of the expression of fatty acid metabolism genes and the abundance of fatty acid showed that the positive correlation between expression of FADS2 and the abundance of C20:5.The expression of FADS1 was positive correlation of the abundance of C20:5 in the tail fat and perirenal fat.The expression of SCD was negative correlation of the abundance of C20:5 and C22:6 in the intramuscular fat.The relative abundance of Firmicutes of the grazed was higher than the concentrated.The relative abundance of Bacteroides of the grazed was higher than the concentrated.The relationship of the expression of the microbial ecology and the abundance of fatty acid showed that the abundance of Fibrobacteres was positive correlation of the abundance of C20:5 in the intramuscular fat and tail fat,and the abundance of Fibrobacteres was positive correlation of the abundance of C22:6 in the intramuscular fat.The abundance of Fibrobacteres was positive correlation of the abundance of C18:1 in the tail fat and perirenal fat.The abundance of Bacteroidetes was positive correlation of the abundance of C20:4 and C22:6 in the perirenal fat,but that was negative correlation of the abundance of Firmicutes.The abundance of Butyrivibrio was negative correlation of the abundance of C18:3,however the abundance of Bacteroides was positive correlation of C18:3.The abundance of Bacteroides was good for the abundance of polyunsaturated fatty acid in the different adipose tissue. | | Keywords/Search Tags: | Sunite Sheep, Feeding method, Fatty acid metabolism genes, Fatty acid, Microbial ecology | PDF Full Text Request | Related items |
