Laminarin is a kind of β-glucan.At present,most of scientific research reports on laminarin are aimed to immunology and anticancer.However,its effects on energy homeostasis of animals were rarely reported.Glucagon-like peptide 1(GLP-1)that secreted from enteroendocrine cells plays an important role on body energy homeostasis via regulating food intake and insulin sensitivity.The study of GLP-1 secretion and its mechanism has a great help on animal energy homeostasis regulation.In order to explore whether laminarin could regulat energy homeostasis through influence GLP-1 synthesis and secretion,we first used intestinal secretin tumor cell line STC-1 as cell model and determined indexes as follows to explore the effects that laminarin on GLP-1 secretion: GLP-1 secretion levels of STC-1 cells;c-Fos protein expression;Intracellar calcium concentration changes.The results showed: during 0-3 h,lamianrin could significantly indced GLP-1 secretion of STC-1 cells at the doses 10 mg/mL.When co-treated the cel s with laminarin and BAPTA-AM,aforementioned effects were reversed.Then,we use C57/BL6 J mice as live models and determined indexes as follows to explore the effects that laminarin on GLP-1 and energy homeostasis in lives: GLP-1 secretions and their relative gene expression in intestine;Gene expression of Adipoctye ssynthesis;Mice body composition;Insulin sensitivity.Results indicated treating C57/BL6 J mice with 1 g/kg of laminarin by intragastric administration could decrease their food intake and blood glucose via promting GLP-1 secretion.We replaced chow diet to high fat diet for chronic experiment and C57/BL6 mice were tragastic administration of saline and laminarin every 2 days.The body weight giain of the mice were decreased by laminarin via inhibiting adpocyties synthesis and relieveing insulin resistance.In summary,laminarin can trigger GLP-1 secretion through increasing intracellular calcium concentration to further affect energy homeostasis.
