| Hericium erinaceus is a kind of large fungus with high nutritive value and unique medicinal value.Extracts from this mushroom have been demonstrated to be effective in treating gastric ulcer,chronic gastritis and other diseases.The flavonoids,one of the main bioactive components found in Hippophae rhamnoides,play an important role in anti-diabetes,antioxidation and protecting the cardiovascular system.H.erinaceus could enrich some trace elements and small molecule compounds in the cultivation process.At present,studies on selenium enrichment of H.erinaceus have been reported.However,to the best of our knowledge,there is no report of H.erinaceus enriching H.rhamnoides flavonoids.In this paper,the cultivation techniques of H.erinaceus enriching H.rhamnoides flavonoids were investigated,which provides scientific basis and technical support for the production of H.erinaceus with high nutrient and high health value.The main results are as follows:1.By measuring dry weight,density and diameter of hyphae in different liquid medium,the optimal liquid medium of H.erinaceus was obtained:soy flour 20g,glucose 20g,wheat bran 10g,and adding water to 1000 ml,pH nature.The dry weight of H.erinaceus mycelium was 1.06g/100mL in this medium,which was significantly higher than that of conventional PD liquid medium and increased by 79.66%.2.By measuring the culture spawn growth rate,yield and fruiting time,the optimal medium formula 5 was obtained:H.rhamnoides leaves 20%,hardwood sawdust 61.5%,wheat bran 15%,soy flour 2%,gypsum 1%and lime powder 0.5%.In this formula,the mycelium growth rate was 6.67mm/d,which was 8.81%higher than that of control.The yield of the fruiting body was 387.53g/bag,which was 12.07%higher than that of that of control.The fruiting time was 86d,which was two days shorter than that of control.3.By measuring flavonoids content,crude protein,total sugar and combined with yield and growth rate in previous step,the best medium for cultivation of H.erinaceus enriching H.rhamnoides flavonoids was obtained:H.rhamnoides leaves 20%,hardwood sawdust 61.5%,wheat bran 15%,soy flour 2%,gypsum 1%and lime powder 0.5%.The content of flavonoids in fruiting body was 742.47mg/100g,which was 37.64%higher than that of control.Additionally,the fat content was significantly higher than the control,and there was no significant difference between other ingredients and control. |
