"Laboratory animals" refers to animals mainly which is used for experimental purposes such as research,teaching,medical treatment,identification,diagnosis,and manufacturing of biological products,such as medicine,pharmacy,biology,and veterinary medicine.Experimental animal principle.In medicine,experimental animals are used to explore the pathogenesis of human diseases and to find ways to prevent and treat them.Mice and rats are the first choice.The subjects of this experiment were Wistar rats from Animal Experiment Center of Jilin University Medical School.The rats produced diarrhea at a weight of170g-220 g,4-5 weeks of age(weeks after weaning),and partially caused death.The recovery took about a week.During the health,the Large female mouse does not produce diarrhea.Due to the cause of diarrhea,experimental animals have a major problem in their growth and development.In this experiment,the rats were housed in cages under the same conditions.When the onset of the disease,Diarrhea male rats,normal male rats and female rats were dissected simultaneously.The main organs and digestive organs were paraffin-embedded and HE staining was performed to observe histology.The results showed that there were bleeding spots in liver tissues of diarrhea rats,presumably due to diarrheal diseases.Samples of duodenal contents and jejunum contents of experimental rats were extracted with DNA,PCR was performed using universal primers,and the products were subjected to 16 S rDNA metagenomic sequencing.The results of the 16 S rDNA sequencing were used for cluster analysis,strain analysis,and Alpha diversity analysis.The clustering results showed that the female had an infection but no symptoms of diarrhea,indicating that the diarrhea type is a male rat-specific disease.The results of microbial diversity analysis and functional gene analysis showed that the number of beneficial bacteriasuch as Lactobacillus and Haemophilus was greatly reduced,including Ruminococcus,Coleobacillus,Gramineae,Bacillus faecalis,Vibrio devulcan,Helicobacter pylori,and Proteus,The number of harmful bacteria such as Bacteroides,Dorea Clostridium and Clostridium XVIII increased,resulting in a disproportionate bacterial population.In this study,16 SrDNAomics technology was used to study the etiology of gender-specific diarrhea in Wistar rats raised by Experimental Animal Center of Jilin University School of Basic Medical Sciences.The results provide etiological basis for the prevention and treatment of diarrheal diseases in rats.At the same time,it laid the foundation for the supply and health protection of experimental rats.
