Effects Of Supplementation Of Different Sea Buckthorn Pomace Levels On Antioxidation And Expression Of Related Genes In Tissues Of Ram | | Posted on:2018-01-05 | Degree:Master | Type:Thesis | | Country:China | Candidate:X Q Han | Full Text:PDF | | GTID:2393330542475214 | Subject:Animal breeding and genetics and breeding | | Abstract/Summary: | PDF Full Text Request | | As the residue of sea buckthorn products,sea buckthorn pomace contains a large number of nutrients and antioxidant substances.Its resources are very rich,while utilization is very low.The purpose of this study was to investigate the effects of dietary sea buckthorn pomace on antioxidation and related gene expression in different tissues of ram,and to demonstrate the suitable range of seabuckthorn pomace in sheep diets.A total of 24 Doporx Small Tail Han Sheep ram lambs(5 month old,25±1 kg)were randomly selected and equally divided into 4 groups with 6 lambs per group,and received diet containing different level of buckthorn pomace(0%,10%,20%,30%).The experimental period was 65 d,and serum was collected at 40 d.At the end of the trial,the samples of testis,epididymis,hepar and kidney were collected and the testis coefficients,the antioxidant index in serum,the mRNA and protein expression of antioxidant in testis and epididymis and the mRNA expression of antioxidant-related gene and enzyme activity in hepar and kidney of ram were detected.The results are as follows:1.Dietary supplementation of sea buckthorn pomace has no significantly effect on the volume and organ coefficient in testis of ram(P>0.05).The enzyme activity of GPx4 and Cu-ZnSOD in serum were significantly higher in 10%group than that in control group(P<0.05)The enzyme activity of SOD in serum was significantly higher in 20%group than that in control group(P<0.05).The enzyme activity of CAT and T-AOC in serum were significantly lower in 30%group than that in control group(P<0.05).The enzyme activity of T-AOC and GSH-Px in serum were significantly lower in 30%group than that in 10%group and 20%group(P<0.05).The enzyme activity of CAT in serum was significantly lower in 30%group than that in 10%group(P<0.05).The results indicated that supplementation of optimal dietary sea buckthorn pomace could improve the antioxidation enzyme activity in serum and had no significantly affected on the volume and organ coefficient in testis of ram.2.The mRNA expression of Cu-ZnSOD in the cauda epididymal was significantly higher in 20%group than that in the control group(P<0.05).The protein expression of Nrf2 in caput epididymal and Cu-ZnSOD in the cauda epididymal were significantly higher in 10%group than that in the 30%group(P<0.05).The mRNA expression of Cu-ZnSOD in caput epididymal and cauda epididymal were significantly lower in 30%group than that in 10%group and 20%group(P<0.05).The mRNA expression of GPx4 in testis and caput epididymal was significantly lower in 30%group than that in control group(P<0.05).The mRNA expression of CAT in corpus epididymal was significantly lower in 30%group than that in 10%group(P<0.05).The results indicated that supplementation of 10%and 20%sea buckthorn pomace could improve the mRNA and protein expression in testis and epididymis of ram,whereas supplementation of 30%sea buckthorn pomace reduced their mRNA and protein expression.3.The mRNA expression of CAT in hepar was significantly higher in 30%group than that in 10%,20%and control groups(P<0.05).The mRNA expression of Cu-ZnSOD in hepar was significantly higher in 30%group than that in control group(P<0.05).The enzyme activity of CAT in hepar was significantly higher in dietary sea buckthorn pomace group than that in control group(P<0.05).The mRNA expression of Cu-ZnSOD in kidney was significantly higher in 10%group than that in 20%and 30%groups(P<0.05).The mRNA expression of Nrj2 in kidney was significantly lower in 30%group than that in control and 10%groups(P<0.05).The enzyme activity of GSH-Px in kidney was significantly lower in 30%group than that in control group(P<0.05).The results indicated that supplementation of 10%and 20%of dietary sea buckthorn pomace could improve the enzyme activity and mRNA expression in hepar and kidney of ram.Supplementation of 30%sea buckthorn pomace reduced enzyme activity and mRNA expression in kidney,while improved the parameters in hepar of ram.In conclusion,supplementation of optimal sea buckthorn pomace(10%and 20%)could improve the antioxidation parameters of ram,whereas supplementation of excessive(30%)sea buckthorn pomace reduced the expression of antioxidation parameters.Therefore,dietary 10%~20%sea buckthorn pomace is optimal. | | Keywords/Search Tags: | Sea buckthorn Pomace, ram, tissus, serum, Antioxidation, gene expression | PDF Full Text Request | Related items |
