The moso bamboo,with the highest ecological and economic value,has a special place in the gramieous plants,and has a broad development prospect.Moso bamboo is native to China,mainly distributes and cultivates in the southern mountains of China.However,dry environment in autumn and winter has a great influence on the yield and quality of moso bamboo.HD-Zip I protein is a kind of transcription factor with the function of abiotic stress response such as drought response.The scientists have identified some HD–Zip I genes functions from arabidopsis,rice,maize and other species but none from moso bamboo.In our study,we first did the bioinformatics analysis and expression patterns analysis to the moso bamboo genome.According to the results,we selected a HD-Zip gene of moso bamboo,PeHDZ45,which strongly indued by drought stress,for further research.Subsequently,we cloned PeHDZ45 from moso bamboo.The analysis of molecular-characteristics of PeHDZ45 included expression patterns analysis,plant tissue expression pattern analysis,subcellular localization analysis and transcriptional activity analysis were followed then.The main results we obtained are as follows:1.A total of 48 non-redundant HD-Zip genes were identified in the moso bamboo genome.The analysis of these genes were involved structure and evolution.Results showed that almost all moso bamboo HD-Zip genes have highly conserved sequences,they were divided into four well-conserved subfamilies(HD-Zip I to IV)based on their phylogenetic relationships.And the HD-Zip genes between moso bamboo and other monocotyledon plants like rice and maize share similar gene structure.2.Gene structure and sequence analysis showed that all moso bamboo HD-Zip genes have a typical HD and Zip domain.3.We cloned PeHDZ45,a drought strongly induced gene from moso bamboo.Sequence analysis showed that the full-length cDNA of PeHDZ45 is 696 bp,which encodes a protein of 231 amino acids.The tissue-specific expression indicated that PeHDZ45 highly expressed in the root of moso bamboo.4.Subcellular localization analysis showed PeHDZ45 protein is a nuclear protein localization.Yeast hybrid assays showed that PeHDZ45 cannot activate itself.These results will provide important theoretical significance and application value in revealing the mechanism of HD-Zip protein involved in drought response,and also breeding new stress-tolerance varieties of moso bamboo.
