| The morphology and structures of the venom apparatus of four parasitic wasps were observed,including Fopius arisanus(Sonan),Fopius vandenboschi(Fullaway),Diachasmimorpha longicaudata(Ashmead)and Psyttalia incisi(Silvestri)[Hymenoptera:Braconidae],which the host was Bactrocera dorsalis(Hendel)(Diptera:Tephritidae).After that,we preliminary analysed and identified the components of venom proteins,by SDS-PAGE and LC-MS/MS.The main results were as follows:1.The venom apparatus of four parasitic wasps were composed of a yellow venom reservoir,transparent venom glands and a venom duct with sting.It was relatively similar to the venom apparatus of F.arisanus and F.vandenboschi.The venom glands had a number of branches like branches forking,basally inflated and closed end with nearly oval.The only difference of two venom reservoirs which were divided into two distinct areas was in size and shape.The former was similar size and shape about the distinct areas and no significant difference by contrast to vary in the other.For the venom reservoir of P.incisi,it was nearly spherical and the venom glands were generally similar to the first two species.The shape of venom reservoir of D.longicaudata was long oval.Its two venom glands were two branches for branches forking and elongated tubular,narrow at the end.2.Through the band analysis of SDS-PAGE gel electrophoresis of venom proteins,the results showed the molecular weight of venom proteins of F.arisanus focused on 15-75 kDa,and the most abundant proteins were 69.282 kDa,29.18 kDa;the molecular weight in venom proteins of F.vandenboschi was mainly from 20 to 70 kDa,and the protein expressions with 85.812 kDa,69.552 kDa,28.941 kDa,13.092 kDa were much higer than others.The results of D.longicaudata on black strains showed the majority of molecular weight ranged from 15 to 60 kDa.Furthermore,there were 6 more abundant proteins bands with 113.577 kDa,54.769 kDa,38.877 kDa,19.421 kDa,15.308 kDa and 10.767 kDa,respectively.While between 25 and 75 kDa was the main area in the venom proteins of D.longicaudata on red strains,5 more abundant protein bands with 127.86 kDa,49.633 kDa,37.822 kDa,30.879 kDa,18.11 kDa included.As to the venom protein of P.incisi,molecular weight was almost in 25-75 kDa and the proteins of 104.924 kDa,74.134 kDa,63.392 kDa,45.147 kDa and 36.469 kDa were more abundant.3.The results of venom proteins of four parasitic wasps by LC-MS/MS showed,the molecular weights of the venom proteins among the four parasitic wasps were mostly less than 100 kDa,as well as lack of macromolecular proteins that the molecular weight was over 150 kDa.On the acid-base properties of venom proteins,the acid proteins were in the majority and the basic proteins were in the minority.Combined with the band analysis of SDS-PAGE in venom proteins,there were preliminary identifications to the components of the venom proteins.For F.arisanus and F.vandenboschi,there were 17 and 15 venom proteins by analysed and identified respectively;in venoms of D.longicaudata,10 and 14 venom proteins were obtained in black strain and red strain,respectively;For P.incisi,it contained 11 venom proteins.In addition,the classifica-tions and functions of the venom proteins are still unclear,and further studies are needed. |
