| ObjectiveTaking Dendrobium Candidum Wall.ex.Lind as the main object of researc h from Rui Shen Biological Technology Co.,Ltd.This paper mainly studied the efficacy components(such as moisture,polysaccharide,mannose,erianin,etc.)of the stems of Dendrobium Candidum Wall.ex.Lind.from different ha rvest time/properties and different cultivation ways.Meanwhile,To study t he volatile components in the floweres of Dendrobium candidum Wall.ex.Lin d.,providing certain reference for the quality control,the buccal tablet s were prepared and the pharmacodynamics research about the qing pharynx function were carried out.MethodsFirst of all,collecting the new stem and the following year old stems of Dendrobium Candidum Wall.ex.Lind,which are from different harvest time/properties and different cultivation methods.and then,detectioning the moisture content of the fresh stems;Phenol-sulfuric acid method was used to detect the polysaccharide content in the fresh stems of Dendrobium Candidum Wall.ex.Lind;Appling the HPLC methods to detect D-Mannose content and Erianin which is antitumor ingredient content.And based on the above indicators,summarizing the differences and the dynamic change trends with them to determine the enterprise best harvest time and cultivation way at the same time.The volatile constituents in flowers of Dendrobium nobile from four cultivars were identified by gas chromatography-mass spectrometry(GC-MS).Meanwhile,Classifing the common composition and comparing the differences to lay a foundation for the research of scented tea;The supplementary materials and its dosages are determined by the single factor investigation method,forming a best prescription craft to preparate the qing pharynx buccal tablets of Dendrobium Candidun Wall.ex.Lind.and study its quality.The last Study is pharmacodynamics research,which incdued the qing pharynx function of Dendrobium Candidum Wall.ex.Lind through the mouse ear swelling and cotton ball implantation in rats testResulis(1)Moisture determination:The moisture content in stems of different herbs are volatility respectively between 64.74%~86.58%、72.61%~86.58%,and the water content of them is low in January and February.(2)The polysaccharide content percentages in the herbs of Conghua mo untain greenhouse shed、mountains、mountain trees and ground greenhouse cu ltivation respectively are between 15.018±1.8862～41.068±1.2767、17.113±1.5811～35.552±0.74241、20.782±1.4838~40.933±0.83044、18.160±1.3548～35.090±1.0092;The D-mannose content percentages of them respectively are 12.948±0.64306、29.048±0.83607、11.703±0.90829～27.225±1.0697、12.203±1.7284～35.168±2.0737、9.0114±0.40735～28.902±1.0317;and with the increasin g of biological age,the above contents will be increased.(3)Determination of erianin components:the Erianin percentage in Den drobium chrysotoxum Lindl.is 0.032%,but Other herbs have low percentage,even no.(4)43 components were characterized from flowers of Dendrobium candi dum Wall.ex.Lind.,representing 70.64%of volatile components;26 component s were characterized from flowers of Dendrobium thysiflorum,representing 95.24%of volatile components.31components were characterized from flower s of Dendrobium aurantiacum Rchb.f.var.denneanum(Kerr.)Z.H.Tsi,representi ng 72.85%of volatile components;18 components were characterized from fl owers of Dendrobium chrysotoxum Lindl,representing 87.61%of volatile com ponents.(5)The buccal tablet preparation:Optimizing the granulation through using the wet granulation tabletting method and the single factor test.The prescription craft of the buccal tablet were the fixed amount of 30%mannitol as the taste masking agent,70%alcohol as the wetting agent,honey as binder,appropriately adding peppermint oil,the Qing pharynx buccal tablet prepared has the indexes including the uniform colour,the weight variation of buccal tablet was within±5%,the hardness was 45～55 N.Brittle broken degree is less than 1%,all of them are reasonable.(6)Pharmacodynamics experiment about the fuction of Qing pharynx:The test of mouse ear swelling and cotton ball implantation in rats experiment have significant differences between group and blank group.(P<0.05)ConclusionThe free water content in stems of Dendrobium Candidum Wall,ex,Lind from four different cultivation way are different and change with the daily and seasonal change,but the same is that the moisture content of them are all lower in winter than summer,and it is highest during January and March;Whatever the cultivation mode of the herbs,the polysaccharide and mannose contents are all increased with the increase of biological age increasing.Among them,The polysaccharide content of Dendrobium Candidum Wall.ex.Lind imitatiing wild mountain trees cultivated in CongHua is higher than the other three cultivation mode,During January and March,it is higher.In the end,combinating with the determination of the moisture,choose January or February as the best harvest time,the Erianin percentage in Dendrobium chrysotoxum Lindl.is 0.032%,but Other herbs have low percentage,even no.This paper reports,for the first time,the composition of volatile components between flowers of Dendrobium candidumWall ex.Lind,and other three cultivars,A total of 75 components were identified from them,Which were mainly made up of alkanes、fatty acids、esters.The content and chemical constituents from different flowers are varied.The buccal tablets prepared by the wet legal system grain meet the requirements of the pharmacopoeia,and the last optimum perscription effectively improve the taste masking and stability problems.The most important thing is that the pharmacodynamics experiment shows that Dendrobium Candidum Wall.ex.Lind.has anti-inflammatory effect. |
