Spine grape(Vitis davidii.Fo?x)is a wild grape species that sourced in China.As the feature accessions of grape resources,the grape berries are full of nutritious and could be used as table or wine making materials.Meanwile,spine grapevine has strong resistance to fungi diseases and the tolerance to high temperature and high humidity.In this study,the wild spine grapevine from Jiangxi Province were used as materials.For identification of genetic relationship the simpie sequence sepeat(SSR)molecular marker technique was used,and to analysis the leaf shape,the classical measure theory and modern morphological were used.The aims of this study was to reveal the genetic diversity of the wild spine grape accessions and provide a theoretical basis for further collection,preservation,development and innovation of wild spine grape germplasms.The main results are as follows:1.A total of 38 pair of SSR primers were screened from 88 primers,which have polymorphism,reproducibility and distribution in chromosomes uniformity.The genetic similarity coefficient(GS)variation was ranged from 0.67 to 1.0,by SSR molecular marker cluster analysis,at the similarity coefficient of 0.7186,the materials were poly into four categories.The principal coordinate analysis indicated that the first three principal coordinate were 86.1% and 3.8% and 1.5%,the cumulative principal coordinate contribution rate was 91.4%.2.The variation coefficient(CV)of 31 leaf metric traits was between 6.99%-54.08%,the average coefficient of variation was 22.41%,the largest coefficient is the angle of petiole,and the aspect ratio was the minimum coefficient.Principal component analysis(PCA)showed that the first three principal components was 32.35%,17.62% and 10.22%,respectively.The genetic distance was ranged from 0.32 to 2.59 within the 695 spine grape materials,at the distance coefficient of 1.7402,all the materials are divided into thre e categories.The results showed that the leaves of wild spine grape accessions have abundant diversities.Two simple metric traits including circularity and aspect ratio of leaf could used to reflect the variation of blade shape,the leaf with little value of circularity have more serrated and deeper petiole sinus.What?s more,the aspect ratio could be explain the ratio of leaf length to leaf width.Through modern morphological methods,including grape leaf contour of fourier descriptor(EFDs)and leaf venation patterns coordinate general procrustes analysis(GPA),provide the contour and venation patterns of variation information of spine grape accessions has been excavated.3.The leaf characteristics were significantly correlated.There is a significant correlation(P < 0.05)within 31 leaf metric characters;principal component scores of the blade contour has significant correlation with the many metric traits;the elements of three different types leaf venation patterns coordinate principal component score exists significant correlation,the venation patterns coordinate principal component scores has significant correlation with many blades metric characters and blade contour principal component scores.4.The SSR molecular markers and leaf measurement traits clustering results showed that the the correlation coefficient is r=-0.39441,the greater genetic similarity coefficient among the spine grape accessions,the little difference distance between the leaf metric traits.
