| Duration of latex flow after is one of the most important factors that decides rubber yield.The top of laticifer wound formating and accumulating plug materials that lead to the rubber tree laticifer stopped flowing latex.For the reason,the mechanism by which the wounded laticifers are plugged has been a fundamentally theoretical barrier that challenges natural rubber production.Ethrel stimulation caused the tapping tecnolgy invention and application that observably increases yield per tapping.Ethrel stimulation increases latex yield which is ascribed to ethrel effect on significantly prolonging the duration of latex flow.The effect provided conveient for resesrch the wounded laticifers plugging mechanism.Based on this results that the ultrastructure observation of the end of the wounded laticifers and the agglutinate experiment of lutoid bursting lead to latex aggregation,which is believed to result in the plugging of the severd laticifer end,the effect of ethrel on latex flow prolongation has long been ascribed to enhanced lutoid stability.However,some facts do not support this view.Because,after tapping the laticifer wounds were not plugged by the rubber partcdle coagula but by a protein-network,together with rubber particle aggregations.The application of ethrel gas in high concentations results in a signifcant increase the bursting index of lutoids and the duration of latex flow.These results indicatthat the mechanism of the wounded laticifers were plugged need further study.In this paper,we investigated in more detail the mechanism by which ethrel sigificantly prolonging the duration of latex flow by means of cytological,biochemical,molecular biology and proteomic techniques.The results as following:1.Ethrel stimulations can promote the accumulation of protein-network at the end ofthe wounded laticifers.2.β-1,3-glucanases,chitinase and hevein genes expression significantly decreasedfrom lutoid by ethrel stimulations.Ethrel stimulation decreased the three kinds of protein content in the rubber tree variety of PR107,but ethrel stimulation increased the three kinds of protein content in the rubber tree variety of RY8-79.3.β-1,3-glucanases,chitinase and hevein content significantly decreased fromC-serum by ethrel stimulations.4.The gene of 44 kDa protein expression significantly increased by ethrelstimulations.Ethrel stimulations can promoted 44 kDa protein combined with rubberparticles.5.The 44 kDa protein could effectively inhibited latex coagulation caused by B-serum,and rubber particle aggregation caused by protein-network and lutoid debris.The results show that the prolongation of latex flow in etrel stimulated rubber tree may not be related to the cehanced lutoid stability,mainly by increased the content of 44 kDa protein and inhibited rubber particle combined with protein-network and lutoid debris. |
