Life originated from the ocean.In the past three decades,marine natural products have caused widespread concern among scientists.In the vast ocean,there are a large number of new compounds beyond imagination,and a variety of secondary metabolites with novel structures and unique mechanisms of action.In modern medicine,marine natural products have opened up new diagnosis and treatment ideas for common human diseases such as tumors and cardiovascular diseases,and have become the basis for the development of new drugs and the main source of research for important lead drugs.In the future,screening and synthesizing nucleoside compounds with higher biological activity has important scientific significance and social value.In this thesis,the synthesis of marine natural nucleoside compound Kipukasin E has been studied and explored.Based on the o-alkynyl benzoate group,it has excellent characteristics such as mild reaction conditions,good stereoselectivity and wide application range.This group was selected as a new type of protecting group,which effectively avoided the intramolecular ester transfer reaction in the synthesis.?-D-tetraacetylribose as a starting material is cheap and readily available.After 10steps of reaction,o-alkynyl benzoate is used as the ortho-directing group to obtain?-type nucleosides stereoselectively.The glycosylation method was used to construct the C-N bond.Finally,the protective group was selectively catalyzed by Ph3PAu TFA to complete the synthesis of the nucleoside compound.Based on the unique?acidity of gold element,which can activate double bonds,especially carbon-carbon triple bonds,resorufin was used as the fluorescent group and o-alkynyl benzoate was used as the recognition group.A specificity was designed and synthesized Fluorescent Probes Recognizing Gold Ions.The specific recognition of Au1+/Au3+was successfully achieved.In the ultraviolet and fluorescence spectra,the fluorescence probe has a higher selectivity for gold ions than other competitive metal ions,and has good anti-interference ability.It not only realizes fluorescence changes with large emission shift?50 nm?,but also Ions show a significant"naked eye"color change,from light yellow to pink,and also have the advantages of low detection limit,high sensitivity,and wide linear range.In addition,the selection of He La cells has initially explored the feasibility of probe bioimaging applications,showing the promising biological application prospects of this fluorescent probe.
