Pleurotus eryngii belongs to Agaricales,Pleurotus and Pleurotus.It is a large edible and medicinal fungus,which is a high-quality protein source.Compared with animal protein,protein from Pleurotus eryngii has less fat and cholesterol content.Polypeptide from Pleurotus eryngii has the activity of ACE inhibition,which can reduce blood pressure and contribute to the treatment of cardiovascular diseases.In recent years,the output of Pleurotus eryngii has been increasing year by year,but the development of the deep processing industry of Pleurotus eryngii is slow.Using Pleurotus eryngii as raw material to extract ACE inhibitory peptide,the price of raw material is low and the extraction rate of peptide is high.At the same time,it can promote the development of the deep processing industry of Pleurotus eryngii.In this study,the extraction of protein from Pleurotus eryngii and the extraction of ACE inhibitory peptide from protein of Pleurotus eryngii by enzyme were optimized,and the antihypertensive activity of ACE inhibitory peptide in vitro was measured.The main results are as follows:(1)Optimization of protein extraction process of Pleurotus eryngii.On the basis of single factor test,the ratio of material to liquid,ultrasonic power,ultrasonic time and the amount of cellulase were selected as the influencing factors,and the protein extraction rate was taken as the index to optimize the response surface of four factors and three levels.The optimal extraction process of protein from Pleurotus eryngii was obtained.The influencing factors of protein extraction rate were: material to liquid ratio > ultrasonic time > cellulase addition amount The optimal extraction conditions were as follows: ratio of material to liquid 1:30g / ml,Ultrasonic Power 400 W,ultrasonic time 20 min,cellulase content 2%.The actual extraction rate was 75.7%,compared with the theoretical extraction rate of 75.1%,and the relative deviation was 0.79%.(2)Optimization of extraction technology of polypeptide from Pleurotus eryngii.First of all,taking the degree of hydrolysis and ACE inhibition rate as indicators,the enzymatic hydrolysis effects of alkaline protease,pepsin,complex protease,trypsin,flavor protease,neutral protease and papain on the protein of Pleurotus eryngii were compared.The results showed that the hydrolysis effect of alkaline protease and complex protease on Pleurotus eryngii was better.The combination of these two enzymes through single factor and response surface The results showed that the ACE inhibitory rate of the hydrolysate increased first and then decreased.The factors influencing the ACE inhibitory activity of the hydrolysate were time > pH > temperature > enzyme dosage.The optimal extraction conditions were as follows: enzyme hydrolysate temperature 50 ?,enzyme hydrolysate pH7.0,enzyme hydrolysate time 60 min,alkaline protease and recovery The ratio of protease was 4:3.The ACE inhibitory activity of the protein of Pleurotus eryngii was 67.1%,compared with the theoretical value of 65.7%,the relative deviation was 2.1%.
