Tumor is one of the most dangerous killers to human's health and life,so the visual diagnosis and precise treatment of tumor cause wider attention of researchers.It is the breakthrough that fluorescence probe techniques can solve such problems.At present,various types of fluorescent probes have been reported,and commercial fluorescent probes are being pervasively applied for many fields.However,they have some disadvantages,such as fluorescence quenching,low fluorescence quantum yield,susceptibility to external environment,high toxicity,poor solubility in water and no targeting.A novel amphiphilic fluorescent probe was designed and synthesized in the dissertation.Specific content was as follows:?1?BOD-L-C5COONHS was synthesized by bromination reaction,oxidation,Suzuki reaction and acylation,with which the raw material were hydroxybenzaldehyde,pyrrole,ethyl 6-bromocapronate,p-methoxybenzeneboronic acid.Through the optical performance test,it showed good fluorescent effect,and its maximum fluorescence emission wavelength was 613 nm.?2?O-carboxymethyl chitosan?O-CMC?was prepared by carboxymethylation of chitosan in alkaline condition.Subsequently,the BOD-L-C5COONHS was bonded onto the-NH2 of O-CMC,and finally the cRGD peptide was bonded.The fluorescence probe,BODIPY—O-CMC—cRGD,with tumor-targeting and water-solubility was prepared.Two“core-shell”structures of nanoparticles were self-assembled by this probe as the carrier with BODIPY2 and Porphyrin.?3?The structure and biological properties of the probe and nanoparticles were tested by infrared spectroscopy,NMR,SEM,fluorescence spectra and confocal microscopy,respectively.The results show that the fluorescent probe is low toxicity.The nanoparticles self-assembled with the fluorescent probe and BODIPY2 are easily swallowed by tumor cells.The nanoparticlesself-assembled with Porphyrin could not be swallowed by tumor because of the rigid structure and large particle size of Porphyrin.If these two dyes substituted with antitumor drugs in the future,the fluorescent probe is expected to be prepared into an antitumor drug for diagnosis and treatment integration,and could have a broad application prospect.
