In recent years,the need for ingredients from natural sources is becoming imperative due to numerous health risk associated with synthetic additives.Plant essential oils are widely used in food,cosmetics,chemical,fragrance and pharmaceutical industries due to their potential bioactivities,and gradually become an essential part of people's lives.However,most plant essential oils are not widely used in practical applications because of its low water solubility and poor stability.Laifeng chrysanthemum,also known as Shennong Chrysanthemum?Dendranthelna indicum L.?,produced in Enshi city of Hubei Province,which is a new variety of chrysanthemum.Because of its complex composition,domestic and foreign scholars have more research on its chemical composition and content,but there are few studies on the application and bioactivity of Laifeng chrysanthemum resources.In this paper,the essential oils of Laifeng chrysanthemum?Dendranthelna indicum L.?were extracted by steam distillation?SD?and supercritical carbon dioxide extraction?SFE?,and its chemical composition,biological activity and microemulsion were studied to provide scientific basis for further exploration of the application and development of Laifeng chrysanthemum oils.The main conclusions were as follows:?1?The essential oils of Dendranthelna indicum were extracted by steam distillation?SD?and supercritical carbon dioxide extraction?SFE?,and the differences in chemical composition of essential oils were analyzed by headspace extraction-gas chromatography--mass spectrometry?HS-GC/MS?.The results showed that 161 components were separated among which 56 were identified,the main components were alpha.-Phellandrene?10.70%?,Linalool?11.90%?by steam distillation.121 components were separated among which 38were identified,the main components were alpha.-Phellandrene?11.91%?,o-Cymene?22.59%?in the supercritical CO2 fluid extraction.?2?The antioxidant capacity of the SD essential oil and SFE essential oil were compared by six antioxidant systems.The results showed that the two kinds of essential oils had a strong antioxidant capacity in concentration dependent manner.The Dendranthelna indicum oil extracted by supercritical CO2 extraction was more efficacious than the one extracted by steam distillation method through scavenging DPPH radical,ABTS radical,the total reductive power,ferric reducing ability of plasma?FRAP?and the reactive oxygen on Hela cells.But in the anti-yolk lipid peroxidation system,the Dendranthelna indicum oil extracted by supercritical supercritical CO2 extraction was less efficacious than the one extracted by steam distillation method.?3?MTT was used to study the antitumor activity of SD essential oil and SFE essential oil on Hela cells.The results showed that SFE essential oil had better antitumor activity in vitro at the same experimental concentration.?4?The micro-emulsion of SFE essential oils used as material,and the effects of surfactant,co-surfactant,the ratio of surfactant with co-surfactant?Km value?and oil phase on micro-emulsion formation were investigated by pseudo-ternary phase diagrams,and antitumor activity of SFE essential oil and SFE essential oil microemulsion were compared on Hela cells in vitro.The results showed that the best formula of SFE essential oil micro-emulsion was polyethylene glycol stearate-15?HS-15?and glycerol as surfactant and co-surfactant,respectively,the Km was 3,and the mixture of ethyl oleate?EO?and SFE essential oil?4:1?is the oil phase.The micro-emulsion system of SFE essential oil was relatively stable.In addition,SFE essential oil micro-emulsion has better antitumor effect than SFE essential oil.
