Adulteration and fake on livestock and poultry meat has a certain universality,to date there is still lacking mature,reliable,cheap and commercially available meat fraud authentication method.Immunoglobulin G(IgG)is a species-specific protein exists in serum and tissue fluids.Few reports taking IgG as target antigen/protein to discriminate meat's species adulteration or fraud.Taking IgG as the target protein/antigen,adulteration of beef and mutton with horse,pig and duck meat were authenticated by Immunological Agarose gel Double-diffusion Test;cross-reactivity i.e.species specificity of anti-IgG serum was observed and evaluated,and strategy to avoid cross reactivity,and detection limit and robustness of the method were studied,therefor the feasibility of discrimination of the authenticity of livestock meat by Immunological approach discussed.Results indicated that,except rabbit anti-duck IgG serum,rabbit anti-horse IgG serum,rabbit anti-pig IgG serum has a certain cross-reaction with and bovine and sheep IgG or their meat extract fluid,this can be avoid by diluting the serum to appropriate titer.Goat anti-horse IgG serum,goat anti-pig IgG serum observed no cross-reaction with bovine,sheep and camel IgG or their meat leaching juice/fluid.This means preparation of anti-target IgG serum on goat,which in one family with cattle and sheep,can avoid cross-reaction.Goat anti-horse IgG serum which prepared in this study,has a titer of 1:32(25),and its cross-reaction titer with donkey meat juice/fluid reaching 1:16(24).This mean it is not suitable to discriminate donkey meat from horse,but can be used to identify donkey meat adulteration in other meat.Beef and mutton samples were mixed with horse,pig and duck meat by 1%,2%,5%,10%and 50%,and detected by immune double diffusion test.As the results,the detection limit for the three adulterated was 2%-5%,the repetition of result was good,it can fulfill the needs of meat species authentication.In general,it is feasible to taking IgG as target antigen,establishing immunological approach to identify meat fraud.Immune double diffusion test can be an easy,reliable and cheap method to meat authentication.
