Microbial agents have the advantages of improving soil properties and fertility,reducing the using of chemical fertilizer,reducing agricultural production cost,and protecting ecological environment.In this study,the selected functional microobial strains which have strong effects of phosphorus,potassium and nitrogen fixation were used to prepared microbial agents.Meanwhile,the water-soluble biomass calcium sulfonate and ammonium sulfonate were used as organic nutrients for the growth of functional microorganisms,and the culture conditions were optimized and verified.And then,using diatomite as adsorption carrierthe,the preparation technology of solid agent was studied for the purpose of developing agriculture microbial agent products which have strong ability to adapt the environment.The main research results are as follows:(1)The growth of edaphon was accelerated with water-soluble bio-sulfonate.Adding the biomass calcium sulfonate into soil for 30 days,the amounts of bacteria increased by 320%,while which was only a 60%increase in the control.In the meantime the relative abundance of soil microorganisms changed significantly,it shows that soil microorganisms have different ability to absorb and utilize the bio-sulfonate.(2)On the base of interbacterial antagonism experiment,three functional microbial strains without antagonism were selected from the microbial strains isolated and preserved in the laboratory,namely P4,MX 16 and B1.P4 strain and MX 16 strain had the ability to dissolve phosphorus and potassium,among which P4 had the ability to dissolve phosphorus,MX 16 had the ability to dissolve potassium,and B1 strain had the ability to fix nitrogen.(3)The response surface method was used to optimize the initial pH value and the formula of bio-sulfonate medium for the three strains.By using the optimized medium,the flask culture concentrations of P4,MX16 and B1 strains reached 6.79times,3.2 times and 2.9 times respectively before the optimization,and the viable count reached 1.18x109 cfu/mL,9.66×108 cfu/mL and 2.3×109 cfu/mL,respectively.(4)On the basis of the optimized formula of P4,M×16 and B1 strains on the bio-sulfonate medium medium,the fermentation culture in 10L fermenter for 48 h showed that the viable bacteria of P4 reached 1x 109 cfu/mL,the viable bacteria of MX 16 reached 8.59 x108 cfu/mL,the viable bacteria of B1 reached 2.3 x 109 cfu/mL,and the spore yield of all the three strains reached 100%.(5)Solid fungicide was prepared with diatomite as carrier.The adsorption rate of diatomite on the biological-based sulfonate medium is over 60%,which can provide effective nutrition support for the growth of microbial agent.The diatomaceous absorbed with the fermentation broth was dried at 80?for 5 hours.Bacterium agent water content were 30%,26%,24%,fungi in the effective number of living bacterium were 1.5 billion/g,1.7 billion/g,1 billion/g,significantly better than farm microbial agents national standards of the national standard.The three microbial agents mentioned above can be used alone or mixed in any proportion according to the needs of agricultural production.
