At present,large-scale,fully automated chemiluminescence diagnostic instruments for biomarker detection have been widely used in many large hospitals due to their efficient working methods and convenient handling.However,there are few reports on the chemiluminescence instruments that can realize the point-of-care testing(POCT)with miniaturization and simple operation.However,in order to effectively achieve graded diagnosis and treatment,it is necessary to rely on the improvement of the diagnostic ability of the primary hospital,and the large-scale diagnostic equipment is not suitable for its application.Therefore,the development of a small,intelligent,simple operation,fast and accurate on-site detection system has significance.With the introduction of microfluidic chip technology in the 1990s,with the corresponding drive and detection devices,the experimental steps in the diagnostic test can be realized on a chip by manipulating the fluid,including reagents.Storage and release,mixing,dilution,reaction,result detection,etc.This makes the entire detection system(chip and instrument)small and highly integrated,and also reduces the professional level of the inspection personnel,which is highly compatible with the development needs of POCT.Aiming at the above situation,this paper designs a small automatic chemiluminescence quantitative detection platform based on microfluidic chip for detecting biomarkers.The following are the main research contents of this topic:(1)According to the chemiluminescence immunoassay process,a microfluidic chip with complete function and suitable for chemiluminescence immunoassay is designed,including reagent storage and release module,liquid flow control module,magnetic bead mixing module and photon detection module;(2)A driving method for flexibly controlling the flow of liquid in the chip by means of a vacuum chuck is proposed,which changes the way that the conventional external drive must be directly connected to the chip,so that the manipulation of the liquid flow is facilitated and the structure is simple;(3)In order to verify the function of the self-developed microfluidic chip,a fully automated chemiluminescence immunoassay platform was built,and the quantitative detection of commercial ferritin was completed on the chip.The experimental results showed that the photon amount and ferritin concentration of the excitation light were collected.The linear correlation between 5.1-1300 ng/mL was 0.969 and the limit of detection was 2.55 ng/mL.At the same time,this paper also used this microfluidic chip system to detect ferritin in clinical serum samples,and the linear correlation of the test results was 0.979.It can be seen that the designed chip is functionally capable of meeting the needs of chemiluminescence immunoassay.
