China's animal husbandry resources and aquatic resources are rich,annual pork and fish production is huge,resulting in a large number of leather and other processing by-products,if the use of these by-products are incomplete,not only result in waste of resources,but also the natural environment will be A serious threat.Collagen peptide is widely used in food,medicine,cosmetics,biomedical materials,Collagen peptide production of China is often in a state of short supply,pertaining to preparation of collagen peptide from Pig Skin and Fish Skin,the country has published a number of patents and research reports.However,the reported methods generally have the following problems: these methods need to go through the fat and miscellaneous protein and then collagenase is obtained by enzymatic hydrolysis of collagen,the process is complex;In order to remove fat and miscellaneous protein,need to add chemical reagents supernumerary,brought serious of the environmental pollution problem;Need to add expensive protease for enzymatic hydrolysis,high production costs.The use of Bacillus subtilis fermentation preparation of pig skin,fish skin collagen peptide can effectively avoid the above problems,the process is simple,does not pollute the environment,low production costs,is worth promoting the preparation of collagen peptide peptide method.In this study,whole-cell biocatalysts were prepared by Bacillus subtilis fermentation to hydrolyze pig skin and skin collagen,and the hydrolysis conditions were optimized by response surface experiments.The experimental results were as follows: the op timum conditions for the hydrolysis of collagen in pig skin were the ratio of solid to liquid is 1:0.8(g/mL),Hydrolysis time 50 h,hydrolysis temperature 45?,enzyme content 2.72 mL,the hydrolysis rate under this condition is 15.23%;the best condition o f hydrolysis of collagen in fish skin were the ratio of solid to liquid is 1: 6.8(g/mL),hydrolysis time 5h,hydrolysis Temperature 45?,the amount of enzyme is 7mL,in this condition the hydrolysis rate is 35.28%.The results showed that the sensory and physical and chemical indexes of the two collagen peptides are within the quality requirements of the enterprise standard QB2732--2005,and the time-of-flight mass spectrometry analysis showed that the molecular weight of pig skin collagen protein peptide was mainly 237,3261,3427,4311 Da,but it was mainly concentrated in 4311 Da.The molecular weight of fish shin collagen protein peptide was 2113,2721,3402 Da,but it was mainly concentrated in 2721 Da The Amino acid automated analyzer showed that the amino acid contents of pig skin collagen peptide and fish skin collagen peptide were 90.56% and 91%,respectively,and the contents of the two collagen peptide peptides were higher,and they contained hydrophobic amino acid content of 20.29% and 32.92%,respectively,and they don't contain cystine and tryptophan amino acid composition and the primary structure of collagen is very consistent.
