Fluorescent molecular probes,which are based on Fluorescence analysis,combining with molecular recognition,can realize molecular and get the corresponding molecular information though the combination of fluorophore and specific receptors.Specially designed fluorescence molecular probes can transform the micro environment change information into optical signals effectively,which is the communication bridge of macrocosm and microcosm.In this thesis,fluorescent probes for mercury,cysteine/homocysteine,and glutathione?GSH?are synthesized respectively.As follows:?1?A new probe CNP,based on 7-amino-4-methyl-coumarin as the fluorophore and alkyne as the high selectivity recongnition unit,was designed and synthesized.Probe CNP has strong fluorescence with the structure of coumarin,but upon the addition of mercury,the fluorescence quenched almost completely.The line range is 1.00×10-8–3.00×10-66 M and the detection limit is as low as 2.70×10-99 M.Probe CNP can be used to detect mercury in water samples with satisfied recovery and on agar gels.These researches indicated that probe CNP holds promise for applications in Hg2+sensing in environmental samples.?2?A new probe BT-Cl,based on aminobenzene thiazole as fluorophore,was designed and synthesized to distinguish between cysteine and homocysteine qualitatively and quantitatively.When the probe BT-Cl reacts with cysteine or homocysteine,“-Cl”in the probe first substitute the sulphur,then cyclization and take off,terminal amino in benzothiazole restored,and the fluorescence recoverd,to meet the purpose of detecting cysteine,homocysteine.The line range is 0.00–5.00×10-44 M,the lowest detection limits are 1.50×10-55 M and 3.70×10-55 M,respectively.?3?A new probe TP-Br,based on triphenylamine as fluorophore,was designed and synthesized to detect glutathione,without the interference of cysteine and homocysteine.When probe TP-Br reacts with glutathione,thiol substitute bromine atoms,fluorescence quenches obviously.But for cysteine or homocysteine,thiol substitute bromine atoms and addition reaction occurs within the aldehyde to ring off,only weak fluorescence quenched.The line range is 0.00–3.00×10-44 M,the lowest detection limit is 4.80×10-66 M.
