Diosgenin is China's unique dioscorea species,which is the main raw material for the production of saponin.With the ever-increasing demand for steroidal drugs,it is more difficult to rely solely on plants as the main raw material to meet the market demand for saponin.The results showed that the endogenous Bacillus licheniformis SYt1,which produced diosgenin from Dioscorea zingiberensis,provided a new way for the production of saponin,but the previous saponin production was far less than the industrial production The need for its fermentation process to be further optimized.In this paper,the fermentation process of SYt1 strain was used to study the synergistic effect of SYt1 strain and the fermentation technology of SYt1 strain.The culture medium of SYt1 strain was studied.Metal ion optimization,and through the design expert software to filter out the main factor,and through the response surface analysis,the optimal conditions for model prediction.And the results are as follows:?1?Optimization of efficiency factorsThe optimized factors were as follows:the optimum concentration of corn steep liquor was 6‰,and the optimum concentration of dextrin was 14,and the optimal concentration of dextrin was 14‰.The results of the screening of the synergistic factors were applied to the fermentation of small cans.Then,the ventilation volume of 5t small tank of SYt1 strain was optimized.The results showed that the higher the amount of ventilation,the higher the saponin production.When the ventilation volume was 5.5 L/min,the yield of saponin reached the maximum,which was 437.36 mg/L,which was 6.28 times of the control group's yield of 69.59 mg/L.?2?Exploring of Feeding fermentationThe results showed that the feed was fed before the end of fermentation,which could effectively delay the fermentation of the end of the fermentation.The saponin concentration after feeding increased to 1456.42mg/L,which was 26%higher than that before feeding.?3?Optimization of metal ionsThe optimal conditions for the prediction of the model were as follows:CuSO4 0.005 g/L,MgSO4 0.509 g/L,H2MoO4 0.003 g/L,the predicted yield of diosgenin production was168.24 mg/L.The yield of the experiment was 167.35 mg/L,and the yield of diosgenin was3.01 times higher than that of the fermentation.
