The evanescent wave fiber fluorescent biosensor and the yeast two-hybrid method were used to detect the estrogenic binding activity of phthalates in this study.The feasibility of the evanescent wave fiber fluorescent biosensor for detection of the estrogenic binding activity of phthalates was determined by comparing the test results.The variation of environmental estrogen in the sewage treatment process was analyzed by detecting the estrogenic concentration in the effluent of each process stage of a sewage treatment plant.The results were listed as follows:1)In the test of estrogen receptor activity,an immune response could occur between E2 and E2 antibodies.The ER after heating was almost completely inactivated and could not be combined with E2.The Ni affinity column did not produce non-specific adsorption to E2.2)In the preservation stability test,the signal obtained from the receptor protein affinity column after 3 months of storage showed little change compared with the initial signal,indicating that the method of preservation of the receptor protein used in this study could maintain receptor protein affinity column activity for a long time.3)The interference of methanol concentration on the test results indicated that the methanol content would not affect the test results when the methanol concentration was low than 10%.The signal value decreased when the methanol concentration was higher than 10%,due to denaturation of the receptor protein at high concentration of methanol.Therefore,the methanol content should be controlled lower than 10%during the detection.4)Among the seven selected PAEs,the estrogenic binding activities of DMP,DEHP,DEP and DOP were very low,which could be neglected,BBP,DBP and DIPP had certain estrogenic binding activities,and their activity order was BBP>DBP>DIPP.The results of the evanescent wave fiber biosensor and the yeast two-hybrid method were similar,confirming the accuracy of the established method.5)The detection of concentration of environmental estrogen in each process of the sewage treatment plant showed that the estradiol equivalent value in the influent was0.152?g·L-1.The estradiol equivalent value of the anoxic,anaerobic and aerobic sections in the three A2O processes were 0.0259?g·L-1,0.037?g·L-1and 0.00118?g·L-1respectively.In addition,there was no detection of estrogen in the effluent of both secondary sedimentation tank and ultra-filtration tank using this method.This indicated that the three-stage A2O process could remove estrogen effectively from sewage.
