China is the mainly country for cultivating oat,in which the production of oat is high and the price is cheap relatively.Oat protein is also a high quality cereal protein,and it has ideal amino acid ratio,which is close to the nutritional mode recommended by FAO/WHO.Researches have indicated that oats have a certain preventive effect on cardiovascular disease,but the preventive effect of oat peptides on cardiovascular disease and its mechanism are still unclear.As the global aging situation becomes severe day by day,the incidence of cardiovascular diseases also gradually increases.Among them,hypertension and hyperlipidemia are typical symptoms of cardiovascular diseases.In this study,oat peptides were obtained by optimized enzymatic method process.Then,the hypolipidemic and anti-hypertensive effect of oat peptides was carried out,which accumulated the theoretical basis for the deep processing of oats and the application of oat peptides.The main conclusions drawn by the research are as following:(1)According to the single factor and response surface test,the optimum conditions for the preparation of oat peptides were Alcalase 2.4L FG is 1.6 mL/L,substrate concentration is 10%,hydrolysis time is 150 min,temperature is maintained at 52°C,pH adjusted and maintained at 9.2.Under these conditions,the degree of hydrolysis of oat protein reached 14.86%,and the yield of oat peptides is 19.02%.(2)The hypolipidemic effect of oat peptides was studied by establishing a hyperlipidemic rat model.The results showed that comparing to the control group the oat peptides could reduce the serum low-density lipoprotein cholesterol(LDL-C)content(P<0.05)and the atherosclerosis index(AI),increase the serum high-density lipoprotein cholesterol(HDL-C)content(P<0.05).It indicated that oat peptides have a certain ability to regulate lipid metabolism disorder,and can improve the bad condition of atherosclerosis in rats.In order to explore the protective effects of oat peptides on cardiovascular,vascular factors levels were also analyzed in this study.The results showed that compared with the control group,at the molecular level,oat peptides could increase the nitric oxide(NO)and inducible nitric oxide synthase(iNOS)level(P<0.05),and decrease the endothelin-1(ET-1)level(P<0.05).At the same time,inflammatory cytokine interleukin-6(IL-6)was significantly increased(P<0.05)and tumor necrosis factor-?(TNF-?)was significantly decreased(P<0.05).All these results indicated that oat peptides had a beneficial regulatory effect on vascular cell related factors.(3)The anti-hypertensive effect of oat peptides was studied by single and long-term intragastric administration in SHRs.Meanwhile,serum biochemical indexes were measured to explore the anti-hypertensive mechanism of oat peptides.The results showed that the systolic blood pressure reached maximum reduction of15.34 mmHg at 4h after oat peptides were intragastrically administered in single intragastric administrationt.In long-term intragastric administration,the systolic blood pressure began to decrease at the 2 week,and the maximum reduction reached to 15.84 mmHg.It suggested that oat peptides have a significant effect on lowering blood pressure.Serum indexes showed that oat peptides could decrease the levels of endothelin-1(ET-1)(P<0.01),tumor necrosis factor-?(TNF-?)(P<0.05),renin(P<0.05),and angiotensin II(ANG II)(P<0.05)and increase bradykinin(BK)(P<0.01)and nitric oxide(NO)levels,which might be the reasons that oat peptides had anti-hypertensive effect.
