With the development of the ethanol industry,alcohol consumption worldwide is increasing dramatically.It is well known that alcohol is bad for either individual health or our society,and this kind of negative influence is increasingly worse recently.Therefore,it is becoming extremely urgent to search for substances which have sober-promoting function against alcohol to reduce the harm.Previously,with the hard working of our group,the hydrolyzates of Pinctada martensii and Oyster meat were found to have anti-alcoholism effect.Pteria penguin is also an economic shellfish which contains abundant proteins,amino-acid,various vitamins and minerals.So this material is good for preparing sober product.The Pteria penguin was used as the research object and the method of biological enzyme technology was used to get the product of enzyme hydrolysis and the mice tests were taken to evaluate the sober effect.The nutrients and functional factors of enzymic hydrolysates were analysis and evaluated.The formula of Pteria penguin on anti-alcoholism was searched and its effect was assessed.The peptide were isolated and purificated and the amino acid sequence of the peptide had higher activity of ADH was detected.The main results are described as below:1,The effection of hydrolysis products of different enzymes to facilitating metabolism were studided and the anti-inebriation and anti-drunk effects of hydrolysis were researched.The results indieated that neutral protease plays a more important role than the other enzymes in preparing products.The enzymatic hydrolysis product of 19.8 degrees fed mice could lower blood alcohol concentration by 32.4%.Compared to the negative control,the tolerance time of enzymatic hydrolysate prolonged and the sober up time of them were shorter,especially the high degree of neutral protease(P<0.01).So the neutral is the best enzyme to hydrolyze.2,The nutrients and functional factors of enzymic hydrolysates were analysised and evaluated.From the results of the research,the enzyme hydrolysis of Pteria penguin was a low-fat and high-protein matter,with the protein 79.3% of weight,while the fat 5.50%.Otherwise,many functional factors were also contained,such as taurine3.83%,leu3.50%,ala3.28%,glycogen6.32% and betaine 4.56%.In the hydrolysate level,the factors had the contributions to sober as follows: betaine stood first among the function factors,with leu,ala,taurine and glycogen followed it in that order.In a word,the hydrosis of Pteria penguin had high nutritions and functions value.3,When some functional supplements of certain concentration were added in the product within a certain range,some interesting phenomena was found.When taurine,betaine or kudzu extract was added it could potentiate the product activity(P < 0.05),whereas,glycogen,alanine or leucine was added it has no significant on the effect(P > 0.05).Through the mixture designed,the optimal formula is: enzyme hydrolysis 65.6%,taurine 2.5%,extract kudzu 10.4% and betaine 21.6%,its can make blood alcohol concentration in mice dropped 43.92% ± 0.03%.The biological activity assessment of formula was investigated.Comparing with the model group,formula of high,middle and low dose group of mice can significantly extend tolerance time and shorten the drunk maintain time(P < 0.05).Besides,the formula can significantly reduce the mice in the serum ethanol concentration and has the remarkable effect(P < 0.05).4,Two weeks of administration,mice acute alcoholic liver damage model was successfully established.Compared with the normal group,ethanol model group of serum alt index and serum lipid levels significantly increased(P < 0.05).The formula and positive control group can inhibit the ratio of serum AST/ALT,CHOL and TG level rising which were inducted by the acute alcohol poisoning.The SOD active was raised and the glutathione(GSH)content also increased meanwhile MDA was descended.From the pathological we can see,the positive control and different doses of formulas in a certain extent can alleviate the liver damage.5,The peptide of Pteria penguin was prepared by neutral protease and the ultra filtration was used to primary purification.When it got through the DEAE cellulose ion exchange chromatography three peaks were collected and the highest active peak can activate the ADH 50%.This peak followed through the Sephadex G-25 chromatography and four peaks were founded.One of them can highlight the ADH active 72.2% and its molecular weight was 3000-200.With the Sephadex G-15 separating,the highest rate of activation can achieve 77.1%.The reverse phase HPLC(RPLC)was used in measure the pure of final peptide and three main peaks was detected.Electro spray ionization mass spectrum results showed that Lys-Glu and Pro-Phe-Arg were contained.
