We studied the nanovesicles formed by the binding of the melanin nanoparticles and the DOX drug wrapped in the platelet membrane in this paper,and modified the RGD peptide on the surface of the nanovesicles to improve its targeting.The immune escape effect and cytotoxicity of nano vesicles were also tested in vitro.Natural melanin nanoparticles with well dispersibility in water,the particle size was 2-3 nm,were prepared from Auricularia auricula by acid leaching method.MNPs has rich aromatic indole and pyrrole ring structure,it can be used for photothermal transformation and combined with DOX by ?-? interaction.Platelet cell membrane was prepared from mouse blood,after modified the RGD peptide on the surface,than loaded melanin nanoparticles and DOX to form vesicular structure RGD-NPVs@MNPs/DOX.The particle size is 85.92 + 6.28 nm.The drug loading capacity of DOX and MNPs was measured to be 7.6±0.8%and 32.7±1.9%,respectively.The surface charge of RGD-NPVs@MNPs/DOX is-23.5±2.6 mV,which is consistent with PM(-24.513.1mV)and RGD-PM(-22.8 + 3.2mV).Experiments showed that RGD-NPVs@MNPs/DOX has good photothermal conversion efficiency and stability.After RGD modification,the CD-47 protein on the surface of platelet membrane still existed,which proved the integrity of the membrane surface structure.The results of immune escape experiments showed that platelet membrane could effectively help RGD-NPVs@MNPs/DOX escape macrophage phagocytosis.The results of DOX release in vitro showed that in the group of RGD-NPVs@MNPs/DOX by irradiating near infrared laser,the release rate of DOX reached 90%in 72 hours,which ensured the high dose of DOX in the tumor tissue.The results of cytotoxicity test in vitro showed that the IC50 value of RGD-NPVs@MNPs/DOX to HUVECs,MDA-MB-231 and MDA-MB-231/ADR cells after 24 hours of incubation was 3.28±0.98,2.62±0.45 and 3.95±0.69 g mL,respectively.The IC50 value was much lower than that of free DOX.
