Rheumatoid arthritis(RA)is a systemic disease that occurs on a global scale.Its incidence is high,and it is basically difficult to cure.It brings great pain and economic difficulties to human life and health.T he development of therapeutic drugs for rheumatoid arthritis has always been the goal of related scientific researchers and pharmaceutical companies.At present,drugs for the treatment of rheumatoid arthritis mainly include anti-inflammatory drugs,anti-rheumatic drugs,hormone drugs,biological agents,traditional chinese medicine and its compound.With the development of biotechnology,bio-targeted therapeutics have become the vane of rheumatoid arthritis.Targeted ther apy is currently the most effective method in many treatment options.This study was conducted through a cell-based pharmacodynamics experiment in vitro on the extracted isolates of aconite root in order to explore and identify potential substances for new rheumatoid arthritis drugs,especially in the molecular targeted therapy for its key considerations.In this paper,ultrasonic-assisted ethanol impregnation of aconite root was used to extract components.T he single factor analysis and orthogonal experiment were used to optimize the extraction process to obtain the optimal extraction method.For the extracted aconite root ingredients,the column chromatographic analysis technology was used in this experiment.T he effective components were separated from th e aconite root by a rapid separation device and six different components were obtained.T his paper is to explore and identify the medicinal ingredients for the treatment of rheumatoid arthritis from aconite root.Based on rheumatoid arthritis fibroblast-like synoviocytes(FLS-RA),a drug discovery tool for cell proliferation,factor and protein expression as an indicator,can be used for pharmacodynamic studies of rheumatoid arthritis drugs in vitro.T his method is aimed at the specific target drug administration.T he preliminary drug efficacy study of complex drugs can not only lay a foundation for subsequent animal experiments,but also lay a foundation for future drug development.In the paper,the cell pharmacodynamics experiments were performed on the six components extracted and separated from aconite root by using cell proliferation and cytokines and protein expression as indicators.Finally,the different effects of FLS-RA with different components were obtained.Among them,there are no lack of effec tive pharmaceutical ingredients that can be used as early materials for drug development.
