Rhizoma Polygonati is used as medicine with root,have invigorating spleen and replenishing qi,kidney tonifying effect.It is the traditional Chinese medicine materials useing tonifying middle-Jiao and qi,"Long-term use,will be stronger"belong to medicine and food dual purpose Chinese Medicine.Modern pharmacological studies have found that the main active components are steroidal saponins and polysaccharides.Traditionally,the use of Rhizoma Polygonati is mostly cooked,it needs nine steaming and nine drying,which can be achieved by repeated processing.At present,the research on the processing of Rhizoma Polygonati is mainly based on the change of polysaccharides,and it is found that the content of polysaccharide is reduced after processing,and hydrolysis into the easy boil out oligosaccharides and monosaccharides,and produce 5-HMF hydroxy furfural(Maillard reaction).However,what kind of changes have taken place in the process of processing about the main active ingredient of steroidal saponins?At present,there are very few research about it.The steroid saponins compounds(Dioscin)and furostanol steroidal saponins compounds(Protodioscin)are the main ingredients steroidal saponins in Polygonatum kingianum Coll.et Hemsl.We put forward the new method of processing,simulated by monomer.Study on Polygonatum kingianum Coll.et Hemsl of the chemical composition of steroidal saponins before and after changes,combined with NMR and MS to ensure the changes of steroidal saponins chemical compositions,for the first time we found the possibility of plant hormones conversion to animal hormones(C-27 steroidal saponins?C-21 pregnane steroidal saponins).This thesis comprises three chapters.The first chapter summarizes the resource of Polygonatum kingianum Coll.et Hemsl and its chemical constituents and bioactivities of steroidal saponins.The second chapter form a system research about the processing change of steroidal saponins.The third chapter,summary and outlook of this dissertation work.By the means of chromatographic separation methods and modem spectroscopy technology,a system research have been made about the steroidal saponins.Thirteen compounds were isolated,there structures are including C-27 steroidal saponins?C-21 pregnane steroidal saponins.After processing,four compounds are first isolated,among them,there are two new compounds.Other compounds are identified as:Dioscin(1);Protodioscin(2);Methylprotodioscin(3);Protogracillin(4);Methyl protogracillin(5);P-sitosterol(6);Trillin(7);Diosgenin(8);Huangjinoside S(9*);Huangjinoside T(10*);3?-[(O-?-D-glucopyranosyl)oxy]-pregna-5,16-dien-20-one(11);3?-[(O-?-L-rhamnopyranosyl-(1?2)-?-D-glucopyranosyl)oxy]-pregna-5,16-dien-20-one(12);Rhamnose(13).
