In this study,the impact of unsaturated fatty acids from rice bran oil to liver hepatocellular carcinoma(HepG2)was measured and preliminary analysis of its mechanism.MTT method was used to determine changes in cells survival rate and growth curve.Inverted microscope and fluorescence microscope were used for observation of the change on cells morphology.Flow cytometry assay was used to investigate the cell cycle.Transwell chamber migration assay was used to measure the ablility of cells migration.Cells Clone experiments investigated the change of cells clone ability.Western blot was used to detect the change of expression of NF-?B in order to researching and exploring anticancer mechanism.The results showed that the inhibition ability of unsaturated fatty acids which were under certain concentrations from rice bran oil to HepG2 was obviously(P<0.05).When the concertration of RBO,OA,LA,LNA reached more than 0.15mmol/L?0.2mmol/L?0.08mmol/L and 0.15mmol/L,the effect of inhibition was evidently.Cell cycle analysis showed that the percentage of G2 phase cells decreased,cell growth blocked in S phase.Result of light microscope showed that cells were polygon,uniform size,clear edge,abundant cytoplasm and had 2-5 nuclears in each cell,and it became smaller cell body,intercellular space increased.Nucleus became blurred,cell membrane cell fell off shrinkage phenomenon in treated groups.The migration experiment results show that the treated groups migration rate is much lower than the control group,the migration inhibitory rate was 32.85±3.24%,25.55±2.56%,41.61±4.08%,47.45 ± 4.60%.The cell clone experiment results showed that the treated groups was significantly reduced clonogenic capacity,even no colonies appeared.Western blot analysis showed unsaturated fatty acid from rice bran oil down-regulated NF-?B expression levels within overall range and promoted NF-?B to translocate into nuclear.Research results shows that unsaturated fatty acids from rice bran oil may inhibit HepG2 proliferation and reduce their migration and cloning capability.The possible mechanism may in turn activate HepG2 cycle arrest-related factors by promoting nuclear translocation of NF-?B expression occured in S phase to make effect in inhibition the proliferation of HepG2.This will further develop the use of rice bran oil that provides research-based data,and provide a reference for finding and developing a new type of natural anti-cancer substances.It also provides the theoretical basis for the application of unsaturated fatty acids from rice bran oil in the food and pharmaceutical area.
