Ginseng is the precious Chinese traditional medicine,it is famous for the effect of prevention for disease and nourishment.The natural activity of ginseng has valuable medicinal properties.ginsenoside is the primary biological activity.Ginsenoside Rd can promote the exudation of adrenal cortex Hormone and Protect Kidney.It can also promote the proliferation of T-cell and improve the activity of NK.In this essay,the main purpose is preparation of ginsenoside Rd.In this paper,we employ a bacterial enzyme that catalyzes ginsenoside into ginsenoside Rd.Some research including the optimum strain,the optimum reaction condition,the isolation of ginsenoside Rd and the bacterial enzyme and the quale of the bacterial enzyme was done.The experiment confirmed that sp.GS3054 is the best strain and the optimum enzyme inducer,flos sophorae is the optimum inductor.The experiment aslo found when sp.GS3054 was cultured in the 2nd medium it yielded the highest ginsenoside Rd.Furthermore,it was shown that 3 days was the best period of enzyme fermentation at 30 ?.The ginsenoside-glycosidase was purified by DEAE-cellulosecolumn.In the end,the pure ginsenoside-glycosidase'smolecular weight was determined by SDS-polyacrylamidegel electrophoresis as 62 KDa.In order to ginsenoside Rd as much as possible,we studied the optimum reaction condition.Through the research we got that the optimum substrate concentration was 0.1%,the fittest pH was 5.0 and the temperature was 30 ?.In the further,we study some inorganic ions including sodion,potassium ion,ferriion,aluminum ion,copper ion,magnesian ion,factor IV.The ferriion had very effect on enzyme reaction and promote ginsenoside Rd to produce other ginsenoside.The magnesian ion and factor IV had action of promotion.The aluminum ion and copper ion had action of promotion and inhibition.Protopanaxdiol was extracted from western ginseng saponins and the yield was 6%.When silica gel chromatograph was applied on separation,9.46g ginsenoside Rd was obtained from 30g western general ginseng saponins,the yield of which was 32.5%.A 4.70g partially pure ginsenoside Rd was obtained together,the yield of which was 15.6%.we adopt the mothod of crystallization to epurate A 1g sample of ginsenoside Rd was then crystallized and get 500mg pure ginsenoside Rd(the purity was 98%)was obtained,the yield of which was 50%.
