Many epidemiological and clinical studies found the different effect of ruminannt trans fatty acids(R-TFA)and industrial trans fatty acids(I-TFA)on health,R-TFA showed negative realtion with vascular disease,however,I-TFA showed positive relation with vascular disease.The mechanism of different effect of the two trans fatty acis on vascular health was not clear.The endothelial cell dysfunction was the initial factor of vascular disease.Therefore,this project studied the effect of two trans fatty acids on cell viability,cell morphology,oxidase activity(SOD,and MDA),LDH leakage rates,effects of NO content and NOs activity,and investigated the fatty acids variety of endothelial cell after treated by the two trans fatty acids through GC analysis,discussed the effect of two trans fatty acids on inflammation of endothelial cell by detecting the mRNA level of ICAM-1?VCAM-1 and IL-6,to obtain the mechanims of different effect of two trans isomers on endothelial cell dysfunction.The obtained results are as follows: The results showed that 9t18:1 and 11t18:1 can reduce the cell survival rate,inhibitory effect of 9t18:1 on proliferation of endothelial cells were stronger than 11t18:1 in 100 ? mol/l and has a significantly difference.Both of trans fatty acids are lead to changes in endothelial cell morphology,cells from normal spindle to round and edge became rough.1.Both of the two trans fatty acids were lead to changes in endothelial cell morphology,cell was deformated from normal shuttle to circular with rough edge.Both 9t18:1 and 11t18:1 could reduce the cell survival rate,and the cell survival rate was reduced more in 9t18:1 group than in 11t18:1 at concentration of 100 ?mol/L,2.Both of the two trans fatty acids increased LDH leakage rate and MDA in endothelial cell,but 11t18:1 group was lower than that in the same concentration of 9t18:1.Both of two trans isomers lowed NO secretion,NOS and SOD activity,and 11t18:1 group was significantly higher than that in the same concentration of 9t18:1.On the whole,The injury of 11 t18:1 to the human umbilical vein endothelial cells is significantly less than 9 t18:1.3.By comparing the effects of 9t18:1,11t18:1 and 9c11 tCLA on inflammation of endothelial cell,we found that ICAM-1,VCAM-1,IL-6 mRNA expressionwas increased significantly higher in 9t18:1 than 11t18:1,and 9c11 tCLA showed obvious inhibition to the inflammatory response.4.11t18:1 could be partially transformed to 9c11t-CLA in endothelial cell.The conversion rate of VA to 9c11t-CLA was 23% in HUVECs,which possibly was one of the reasons for differnt effect of two trans fatty acids on endothelial cell function.
