The cyanide-resistant respiratory pathway is an important feature distinguishing plant mitochondria from animal mitochondria.It has been proved that cyanide-resistant respiration has important significance in enhancing plant stress resistance.This paper provides a basis for exploring the adaptation mechanism of plants to karst environment by studying the effects of exogenous calcium on the physiological characteristics of the genus Paphiopedilum and the expression pattern of the Alternaria oxidase gene family under environmental stress.To understand the mechanism of karst vegetation restoration succession and provide scientific basis for screening karst suitable plants for seedling afforestation.In this experiment,it was found that when 200 mM CaCl2 was used to treat the Primula rupestris,the chlorophyll content in the leaves of the Primula rupestris was significantly decreased with the prolongation of the treatment time;The net photosynthetic rate,transpiration rate,intercellular CO2 concentration and stomatal conductance decreased sharply,which had adverse effects on plant growth;The degree of membrane lipid peroxidation and cell membrane permeability increased significantly,causing serious damage to cells,and the activity of antioxidant enzymes also changed significantly.The activities of SOD,POD and APX were significantly increased,while the activity of CAT was decreased.Plants increase their resistance to stress by increasing theactivity of antioxidant enzymes when subjected to stress.In summary,excessive concentrations of exogenous calcium can cause damage to plants and are not conducive to plant growth.Based on the transcriptome data of the Primula rupestris,the Blast gene was sequenced with the AOX family gene sequence of Arabidopsis thaliana,three genes PrAOX1 a,PrAOX1d and Pr AOX2 were identified.The expression specificity of AOX gene family members under different treatment conditions was explored by high-throughput sequencing platform and qPCR.The results showed that under stress conditions,PrAOX1 a gene expression was up-regulated,while PrAOX1 d gene expression was down-regulated,and PrAOX1 a and PrAOX1 d was induced to achieve the highest expression level when treated with 200 m M CaCl2 at 7 day,Pr AOX2 was not induced,and PrAOX1 a was more up-regulated by stress-induced genes.A gene that regulates the resistance of the Primula rupestris.The high calcium environment has an adverse effect on the growth and development of plants,and the different genes of the AOX gene family have different expression characteristics,which jointly regulate the cyanide-resistant respiration of plants.
