Flowers are important organs of plants and are regulated by a variety of flower development factors.APETALA 1(API),AP2,AP3,PISTILLATA(PI)and AGAMOUS(AG)are important genes affecting flower development.EMBRYONIC FLOWER 2(EMF2)and VERNALIZATION 2(VRN2)are one of the subunits of the Polycomb Repressive Complex 2(PRC2),and the emf2-10 vrn2-1(her)floral organs are significantly different from the wild type.In this study uses Arabidopsis thaliana as experimental material,the emf2-10 vrn2-1(Ler)double mutant is corssing with apl(Ler),ap2(Ler),ap3(Ler),pi(Ler)and ag(Ler)single mutants,we get triple mutant by phenotyping.Using dissecting microscope to observe and record the abnormal development of wild-type,single-mutant,double-mutant and triple-mutant floral organs,and to compare the differences between mutant plants and wild-type floral organs in combination with the flower development model,to explore the regulation of Ploycomb Group proteins in the determination of floral organ identity.The qRT-PCR assay was used to compare the expression differences of API,AP2,APS,PI and AG in Arabidopsis wild type and emf2-10 vrn2-](Ler),and further explore the epigenetic regulation mechanism of Polycomb Group proteins on flower organ identity.The experimental results are as follows:First,the emf2-10 vrn2-1(Ler)double mutant is corssing with ap1-1(Ler),ap2-7(Ler),ap3-1(Ler),pi-1(Ler)and ag-1(Ler)single mutants,we get triple mutant.Secondly,select Ler,ap1-1(Ler),ap2-1(Ler),ap2-7(Ler),ap3-1(Ler),pi-1(Ler),emf2-10 vrn2-1(Ler),apl-1 emf2-10 vrn2-1(Ler),ap2-1 emf2-10 vrn2-1(Ler),ap2-7 emf2-10 vrn2-1(Ler),ap3-1 emf2-10 vrn2-1(Ler)and pi-1 emf2-10 vrn2-1(Ler)under the same growth conditions,performing floral organ morphology observation,the results of flower organ morphology analysis showed that the Polycomb Group proteins have the function of clear floral organ ideantity.Finally,through qRT-PCR analysis of genes in Ler and emf2-10 vrn2-1(Ler),it was found that API,AP2,AP3,PI,AG and SEP3 all had a decreasing trend,while LEAFY(LFY)and TERMINAL FLOWER 1(TFL1)had an increasing trend,indicating that the epigenetic regulatory factor PRC2 plays an important role in the development of plant floral organs.This paper lays the foundation for the study of epigenetic mechanisms in flower development.
