The static magnetic field has been widely used in science due to its constant intensity and less parameter changes. For example, the development of nuclear magnetic resonance and strong static magnetic field technology has greatly promoted the development of biology and physics. As its application increasing, the health risks are also receiving more attention. The reproductive system is an important part of the biological reproduction. The germ cells are responsible for the transmission of genetic information. In recent years, there have been more studies on the germ cell effects of the static magnetic field, but the conclusions and mechanisms are still not clear. In this study, the effects of static magnetic field on germ cells were studied with C.elegans as a model organism, and its mechanism was further studied. The results are summarized as follows:1.The effect of SMF on the spermatogenesis of male C.elegans After male C.elegans exposure to 10 T SMF,we found that SMF treatment had no significant effect on the activation of sperm, but the SMF caused an increase in the early activation of sperm. Then,we tested the transfer of sperm and the brood size respectively, and found that the brood size of males decreased after exposure to SMF,and the number of males that could not transfer sperm increased;however, the migration of male sperm in females had no significant change.2.Mechanism of SMF-induced increase of germ cell apoptosis We found that after exposure to 10 T SMF,the germline apoptosis of C.elegans was significantly increased. After co-exposure with the iron chelator DFO,the phenomenon of increased germline apoptosis was significantly alleviated. Then,we separately detected the free iron content and the expression of ferritin in the C.elegans.We found that compared with the control group, the free irons in the C.elegans increased significantly after exposure to SMF and interestingly, the expression of FTN-1 was reduced significantly, but the expression of FTN-2 was not changed significantly. This proved that iron metabolism played an important role in increasing the germline apoptosis, induced by SMF.Conclusion:The 10 T static magnetic field induces the early activation of spermatozoa of C.elegans, reduces the transfer of male sperm, and increases the apoptosis of germline cells by interfering with the metabolic balance of iron,it provides a new experimental basis for clarifying the reproductive effect of magnetic field.
