At this stage,PCV2 is prevalent in the world and poses a great threat to the healthy development of pig industry.It is not only the pathogen of weanling piglet multi-system failure syndrome,but also causes porcine reproductive disease,leading to PCV2-related pneumonia and related enteritis.At present,the problem of diarrhea caused by PCV2 is very severe in China,the result of loss of appetite,weight loss problems generally plagued the farmers and brought huge economic losses.Find out how PCV2 causes piglets to diarrhea is an important way to avoid and treat diarrhea.In the body's intestinal tract,the stability of its internal environment and intestinal mucosal immunity are closely related,so this article on the impact of PCV2 on intestinal mucosal immunity made the following aspects of the study.The first part was to investigate the effect of PCV2 on intestinal SIgA and related cytokine levels.PCV2 was used to infect 5-week-old Kunming mice.The mice were treated with PCV2 on day 7,day 14,day 21,day 28,Day and day 42,mice were sacrificed by inguinal lymph nodes,spleen and Peyer's knot.The virus was inoculated into the cells for identification.Also,the protein was detected.Then,is extract SIgA and several characteristic cytokines such as IL-2,IL-4,IL-10 and IFN-? and test analysis.The results showed that the levels of SIgA,IL-2 and IFN-? in the intestine of the test group increased first and then decreased with the prolongation of infection time,and the levels of IL-4 and IL-10 were increasing,The results showed that the contents of SIgA and related cytokines in mice were changed under the stimulation of PCV2,which affected the level of intestinal immunity and the disorder of the intestinal level.Road normal physiological function damage.The second part is the study of the correlation between PCV2 viral load and pathological damage.In this study,we used PCV2-infected mice to establish a qPCR technique and paraffin using SYBR Green I chimeric fluorescence method.The HE staining method was used to detect the virus by qPCR at 7 days,14 days,21 days,28 days,35 days and 42 days after infection.The virus was quantitatively analyzed,Tissue for the production of slices,staining observed intestinal pathological changes.The results showed that the Peyer's nodules of mouse intestinal Peyer's nodules showed necrosis and deletion of lymphocytes.When mild infection,the nucleus was dissolved,the area of nuclear rupture was small,and the cell was absent.In the severe case,the cytoplasm was dissolved and the nuclear rupture,Nuclear dissolution phenomenon,in the first 14 days,21 days high viral content,lymphocytes also showed more serious necrosis.
