Objective: To investigate the method of inducing the transdifferentiation from fibroblasts to oligodendrocyte precursor cells(OPCs)by micro RNAs.Methods: The fibroblasts derived from newborn SD rat skin were isolated and cultured.After identified through microscope and immunofluorescence,the cells were transdifferentiated with mi R-219 and mi R-338.Then the cells were cultured in the OPCs inducing medium.The specific OPCs markers of O4,Olig2,and A2B5,as well as the oligodendrocyte(OLs)marker of A2B5,were detected via the immunofluorescence technology at the 2ed day,4th day,6th day,8th day and 12 th day.Besides,RNAs were extracted for detecting the expression of Olig2,O4,MBP,MAG,MOG,GFAP,Fibronectin.Results: After fibroblasts were transfected with mi R-219 and mi R-338 for 4 days,the morphology of the induced cells began to change from typical flat to latticed.Immunofluorescence result showed Olig2,O4,MBP were positive,but A2B5 was negative.RT-q PCR results showed the expression of Fibronectin was in an increasing trend,While the expression of OPCs markers were in an increasing trend at first,then decreased during the last few days.The markers of OPCs and OLs in the negative control group and blank group were not detected.Conclusion: The fibroblasts were transdifferentiated into OPCs-like cells via mature mi R-219 and mi R-338 in vitro.
