1 ObjectiveTo study the effects of regular exercise on the cardiac function,sleep and climbing ability in drosophila with cardiac-specific dFoxO gene overexpression at young age,and explore the anti-aging function of exercise of dFoxO.2 MethodsHand-Gal4 males were crossed to W1118 or UAS-dFoxO overexpression virgin females and incubated at temperature of 25?throughout development.Then the adult F1 progeny were collected within 6h after eclosion and respectively divided into 8 groups.There were Hand-Gal4>W1118–3WC,Hand-Gal4>W1118-3WE,Hand-Gal4>UAS-dFoxO-3WC,Hand-Gal4>UAS-dFoxO–3WE,Hand-Gal4>W1118–5WC,Hand-Gal4>W1118-5WE,Hand-Gal4>UAS-dFoxO-5WC,Hand-Gal4>UAS-dFoxO–5WE.Each group were 400.All flies were kept at 25?and 50%-60%humidity,on a 12h lighdt/dark cycle,and transferred to fresh food once every2 days.The flies in the exercise groups at 2 days old performed platform exercise 2 hours per day for 3 or 5 weeks.Exercise training was performed at 9:00-11:00.The dFoxO mRNA expression in cardiac muscle was measured by Real time qPCR and heart function was recorded by M-mode echocardiography.Activity was measured using the Drosophila Activity Monitoring System?DAMS?.All test was completed after the period of exercise training.3 Results3.1 The result of dFoxO mRNA expression in cardiac muscleThe expression of dFoxO in 1 week old UAS-dFoxO control group was increased by 56.7%compared to 1 week old control group.It was demonstrated that overexpression of dFoxO was constructed successfully.3.2 The result of heart function of fruit flies by M-modeCompared with Hand-Gal4>W1118–3WC group,diastolic diameter,fractional shortening and fibrillation of Hand-Gal4>UAS-dFoxO-3WC group decreased?P<0.05?.By contrast with Hand-Gal4>UAS-dFoxO-3WC group,diastolic diameter,fractional shortening in Hand-Gal4>UAS-dFoxO-3WE group was significantly increased?P<0.01?.Compared with Hand-Gal4>W1118–3WE group,fibrillation was significantly decreased?P<0.001?in Gal4>UAS-dFoxO-3WE group.Compared with Hand-Gal4>UAS-dFoxO-5WC group,diastolic diameter,fractional shortening of Hand-Gal4>UAS-dFoxO-5WE group decreased?P<0.05?.By contrast with Hand-Gal4>W1118–5WC group,fibrillation was significantly decreased in Hand-Gal4>UAS-dFoxO-5WC group?P<0.05?.3.3 The result of sleep of fruit flies by DAMSCompared with Hand-Gal4>UAS-dFoxO-3WC group,evening sleep episode duration and evening activity increased obviously?P<0.01?,evening activity and total activity decreased significantly in Hand-Gal4>UAS-dFoxO-3WE group.Compared with Hand-Gal4>W1118–3WE group,total sleep of Hand-Gal4>UAS-dFoxO-3WE increased obviously.?P<0.01??Compared with Hand-Gal4>W1118–5WC,evening sleep episode duration and total sleep increased in Hand-Gal4>W1118–5WE group?P<0.05?.Compared with Hand-Gal4>UAS-dFoxO-5WC group,evening sleep episode duration increased obviously?P<0.05?,evening activity decreased?P<0.05?in Hand-Gal4>UAS-dFoxO-5WE.By contrast with Hand-Gal4>W1118–5WC group,daily bout number and evening bout number decreased in Hand-Gal4>UAS-dFoxO-5WC group?P<0.01?.3.4 The result of climbing index across agesThe exercise groups displayed decreased climbing index across ages.Compared with Hand-Gal4>W1118–5WC,climbing index index increased obviously in Hand-Gal4>UAS-dFoxO-5WC group?P<0.05?.4 Conclusion4.1 dFoxO plays an important role in maintain cardiac function,sleep and mobility.dFoxO gene overexpression decreased fibrillation and climbing index in aging flies.4.2 Exercise in early age can improve fractional shortening and sleep quality caused by dFoxO gene overexpression.
