The basic research of Ying Xin Dan is very few,the existing research is so long that there is no basic research on Ying Xin Dan in the near future.Moreover,it is not clear whether Ying Xin Dan is effective for animal models of acute coronary heart disease and myocardial infarction,or for animal models similar to chronic coronary heart disease,and its pharmacodynamic action is also not clear about its prevention or treatment.The mechanism of action is also not clear.The research work includes three parts:the first part is the research of the preventive protection Ying Xin Dan function and mechanism of the approximate model of variant angina animal,and the second part is the effect and mechanism research of Ying Xin Dan on the approximate acute myocardial infarction animal model,and the third part is the effect and mechanism of Ying Xin Dan on the approximate unstable angina pectoris animal model.The first part the research of the preventive protection Ying Xin Dan function and mechanism of the approximate model of variant angina animalFirst:Preventive protective effect and mechanism of Ying Xin Dan on myocardial ischemia model of pituitrin(pit)rats injected under hypoglossal veinA rat model of myocardial ischemia induced by sublingual intravenous injection of pit was established to simulate the variant angina pectoris,and to further explore the effect and mechanism of Ying Xin Dan on the similar variant angina pectoris.The results showed that Ying Xin Dan could reduce the electrocardiogram ST segment elevation and decrease the arrhythmia rate of pit induced acute myocardial ischemia in rats,which had a good effect on the early stage of myocardial injury,and could also improve the pathological damage of myocardium.Ying Xin Dan on pit induced similar variant angina had protective effects on acute myocardial ischemia in rats,which the best was the large dose(four times of clinical dosage)of Ying Xin Dan;its mechanism may be related to coronary spasm,improve myocardial ischemia antagonist contraction function,antioxidation and reduce myocardial damage.The second part The effect and mechanism research of Ying Xin Dan on the approximate acute myocardial infarction animal modelFirst:The preventive protective effect and mechanism of Ying Xin Dan on myocardial ischemia model of rabbits with intravenous injection of isoproterenol hydrochlorideThe experimental results showed that ISO could improve Ying Xin Dan caused myocardial ischemia in the rabbit heart swelling degree and the ECG ST segment and T wave and heart rate of pathological changes,reduce the content of myocardial injury enzyme,had protective effects on myocardial ischemia in rabbits,and its mechanism may be related to increasing the body’s ability to scavenge free radicals and inhibit inflammatory factors.Second:The preventive protective effect and mechanism of Ying Xin Dan on myocardial ischemia model in rats with coronary artery ligationThe results of the study showed that Ying Xin Dan could improve heart arrhythmia caused by coronary artery ligation in rats with myocardial ischemia,improve the survival rate and improve the ECG S-T segment(J)、T wave and heart rate of pathological changes,reduce the content of myocardial injury enzyme,have protective effects on myocardial ischemia in rats,and its mechanism may be weakened in vivo lipid peroxidation,enhance the ability to scavenge free radicals and inhibit inflammatory factors.In this study,we established two models to simulate acute myocardial infarction in coronary heart disease(CHD)to explore the effect and mechanism of Batan on acute myocardial infarction.The results showed that Ying Xin Dan could improve the myocardial necrosis area caused by myocardial ischemia,effectively improve the pathological changes of ECG,reduce the leakage of myocardial enzymes,and have a preventive effect on the myocardium.The high(four times of clinical dosage)and middle dose(two times of clinical dosage)of Ying Xin Dan is better.Therefore,it can be concluded that Ying Xin Dan had protective effect on the approximation,an animal model of acute myocardial infarction and its mechanism:Ying Xin Dan could increase antioxidant enzyme activity,reduce lipid peroxidation end product to maintain the oxidant/antioxidant balance system tends to a steady state,have anti lipid peroxidation;Ying Xin Dan could decrease the level of inflammatory factors and play a role in protecting myocardial injury.The third part The effect and mechanism of Ying Xin Dan on the approximate unstable angina pectoris animal modelFirst:Study on the effect and mechanism of Ying Xin Dan on carotid artery balloon injuried+ high fat diet and intraperitoneal injection of Chinese spotted viper venom(CRVV)and atherosclerosis histamine triggered rabbit modelThis study established the carotid artery balloon injuried + high fat diet and intraperitoneal injection of Chinese spotted viper venom(CRVV)and atherosclerosis histamine triggered rabbit model is close to the process of unstable angina pectoris.The study found that Ying Xin Dan could stabilize vulnerable plaque,reduce the mortality of atherosclerotic plaque rupture,and reduce the rate of arterial embolism caused by plaque rupture.The mechanism is:Ying Xin Dan could reduce the level of blood lipid,relieve oxidative stress,maintain the balance of oxidation/antioxidant system,inhibit the release of inflammatory factors,reduce the inflammatory reaction in plaque,and prevent the excessive degradation of matrix in plaque.
