Objective:In this experiment,we compared the effect of sham operation group,electroacupuncture group,needle knife group and drug group on Parkinson’s disease model rats.The behavior,morphology and tyrosine hydroxylase positive cell expression,striatum neurotransmitter dopamine and norepinephrine in rats were observed under different intervention modes.The changes of the content of serotonin and the effect of needle-knife intervention on Parkinson’s disease model rats were explored.Methods:The use of SPF SD male rats 96,randomly grouped into blank,sham surgery,model,electroacupuncture,needle knife and drug 6 groups,16 in each group,using six hydroxy dopamine right striatum double target injection method to prepare Parkinson’s disease rat model,through intraperitoneal inj ection of morphine after the induction of circular behavior observed behavioral changes.During the experiment,the blank group,sham operation group and model group did not intervene,and the electroacupuncture group selected "Baihui" and"Taiyang" electric needle 20min,3 times a week,4 consecutive weeks.The needle knife group to take the first,second cervical spine transverse process for loosening,2 times a week,consecutive 4 weeks.Drug group,with the madopar in accordance with the 5mg/100g dosage of the suspension,for rats to fill the stomach,1 times a day,4 consecutive weeks.4 weeks after the end of the intervention period,the changes of the content of the rats ’ behavior,black morphology,striatum neurotransmitter dopamine,norepinephrine and serotonin were observed.Results:Different interventions on behavior of Parkinson’s disease model rats.The results are visible,compared to the blank group,the model group,electric needle,needle knife,drug four groups of circle number are greater than equal to 7 cycles,significantly more than the blank group(p<0.01),suggesting the success of the model preparation.The sham operation group and the blank group compare,did not see the statistical difference,does not appear the circle(p>0.05),suggests the experiment results were stable.Compared with the model group,electric needles,needle knife,drug three groups of circles than the model group significantly reduced(p<0.01),suggesting that electroacupuncture,needle knife,drug intervention can reduce Parkinson’s disease model of the behavioral symptoms of rats,but the electric needle,needle knife,drug group 22 compared,no statistical differences(p>0.05).It was suggested that three groups of intervention methods had an improved effect on the behavior of the rats with Parkinson’s disease,but the differences were not significant.Different interventions on morphology of midbrain substantia nigra in rat model of Parki nson’s disease.The morphological changes of the rat model of Parkinson’s disease were observed under light microscope,there was no obvious difference between the blank group and the sham operation group,the number of neurons was more,the cell was arranged and the gap was small,the cytoplasm cytoplasm was even,the cell size was moderate,the number of glial fibers was small,the number of glia was very low,and the structure of the cells was obvious,the cell nucleus migration,the staining becomes shallow,the intercellular gap widens with the irregular hyperplasia of the glial fiber.Electroacupuncture,needle knife,drug intervention,see the number of neurons in the black area increased,the arrangement tends to be regular,cell morphology than the model group changed significantly,the nucleus is centered,cytoplasm staining deepened,the cell body began to grow larger,while the glial line for hyperplasia there is remission.The arrangement is regular in the model group,which indicates that three kinds of intervention can cause the change of the morphology of the black neurons.Observation of the positive cell of the black tyrosine hydroxylase under immunohistochemical microscope.The blank group and the sham operation group showed that the concentration of tyrosine hydroxylase positive cells in the black area was many,and there were a large number of cell bodies with dense and orderly distribution,and the staining was deep.In the model group,several tyrosine hydroxylase positive cells dispersed in the black area were found,the cell bodies were few and the number of the surrounding protrusions decreased,the distribution was messy and the staining was light.The number of tyrosine hydroxylase positive cells in electroacupuncture,needle knife and drug were more obvious than that in the model group,and the staining was more intense.Overall,the distribution range of the black tyrosine hydroxylase positive cells in the needle knife group was larger than that of the electroacupuncture group and the drug group.Different interventions on the neurotransmitter of striatum in rat modelof Parkinson’s dis ease.①Comparison of dopamine content in striatum of rat 6 group.Compared with the blank group,the content of dopamine in the three groups of sham surgery,needle knife and drug was not statistically significant(p>0.05),and the dopamine content in the model group was significantly decreased(p<0.01),the content of dopamine in electroacupuncture group was lower than that in blank group(p<0.05).Compared with model group,the dopamine content in Electroacupuncture group was increased but there was no statistically significant difference(p>0.05),and the dopamine content in needle knife and drug group was higher than that in model group(p<0.01).Compared with electroacupuncture group,the dopamine content in drug group was higher than that in electroacupuncture group(p<0.05).Compared with drug group,the dopamine content of needle knife group was lower than that in drug group(p<0.05).②Comparison of norepinephrine content in rat striatum of 6 Group.Compared to blank groups,the content of norepinephrine in the three groups of sham operation,electroacupuncture and drugs was not statistically significant(p>0.05),and the content of norepinephrine in the model group was significantly decreased(p<0.01).Compared with model group,the content of norepinephrine in electroacupuncture group and drug group was higher than that in model group(p<0.05).Compared with electroacupuncture group,the content of norepinephrine in needle knife group and drug group was elevated,but there was no statistically significant difference(p>0.05).Compared with drug group,the norepinephrine content of needle knife group was lower than that of drug group,but the difference was not statistically significant(p>0.05).③Comparison of the contents of 6 in rat striatum.Compared with the blank group,no statistically significant differences were found in the content of serotonin in sham surgery,electroacupuncture,needle knife and drug.The content of serotonin in model group was significantly decreased(p<0.01).Compared with model group,the content of serotonin in electroacupuncture group and drug group was higher than that in model group(p<0.05),and serotonin in needle knife group increased significantly(p<0.01).Compared with electroacupuncture group,the content of serotonin in needle knife group and drug group was higher,but no statistically significant difference(p>0.05)was found.Compared with drug group,the content of serotonin in needle knife group was not statistically significant(p>0.05).Conclusion:Needle knife intervention can alleviate symptoms of Parkinson’s disease model rats behavior,promote Parkinson’s disease model rat midbrain black nigra recovery,promote the striatum neurotransmitter dopamine,norepinephrine and serotonin levels increase,to realize the treatment of Parkinson’s disease.
