Objective:Using the international general line of the brain artery embolism method(MCAO)to establish the model of focal cerebral ischemia in rats,the inflammatory factor and blood rheology and angiogenesis for the detection of the core application of molecular biology technology related indicators,combined with the traditional Tibetan medicine theory,explore the medical treatment of Bai Mai therapy(comprehensive therapy using Ruyi Zhen Bao pill and Bai Mai ointment combined)for ischemic cerebrovascular disease,in order to elucidate the mechanism of Tibetan Bai Mai therapy in ischemic stroke rats to provide experimental basis for.Methods:(1)Modeling methods and grouping:according to Belayev’s report,a modified middle cerebral artery occlusion(MCAO)model was established to improve the right middle cerebral artery occlusion artery.The Longa score five was used to score the score.The rats were randomly divided into six groups:sham operation group,model group,Bai Mai therapy group,positive control group,Bai Mai ointment group and Ruyi Zhen Bao pill group.Each group was divided into four subgroups according to ischemia reperfusion time,namely 1 days,3 days,7 days and 14 days.Inflammatory factors appeared in the acute phase.Therefore,1 days and 3 days were selected.The blood vessel regeneration was mostly concentrated in the chronic recovery period,and the time was 7 days and 14 days.Each group was filled with stomach every day,and the medicine was applied to the affected limbs.The positive control group was intragastrically administrated with simvastatin,Ruyi Zhen Bao Wan and the Bai Mai treatment group,and the rest group was intragastrically administrated with normal saline.The rats in the Bai Mai ointment group and the Bai Mai group were smeared with the Bai Mai ointment on the affected limb,and the other groups were coated with vaseline.Detection indexes:① Physiological and pathological effects of brain tissue:HE staining was used to observe inflammatory cell infiltration in hippocampus and cortex,And nuclear condensation and softening of neurons in the hippocampus.② Behavioral examination:behavioral scores of rats in each group were scored on 1 days and 3 days,respectively,including postural reflex test and limb placement test.③Blood rheology test:blood viscosity,plasma viscosity and red blood cell index were detected by hemorheological technique.④Detection of inflammatory factors:the dynamic changes of interleukin-1 beta and tumor necrosis factor alpha in cerebral cortex and blood were detected by Elisa and PCR.⑤Detection of microvascular regeneration and apoptosis:the dynamic changes of CD34 in the cortex of rats were detected by immunohistochemistry.Results:(1)The HE staining showed that the images of the model group showed a clear ischemic core and penumbra,and the brain tissue was loosely arranged and scattered,showing a softening lesion and the infiltration of inflammatory cells in the cerebral cortex and hippocampus of the rats in the white pulse therapy group.The morphological changes of the nucleus and the degree of the softening range were lower than those in the model group.(2)Behavioral detection:two behavioral studies showed that the limb function of the rats in the Bai Mai therapy group was better than that in the model group,and there was a significant difference in three days(P<0.05).The body function of rats in 3 days was better than that of 1 days.(3)Blood rheology detection:Bai Mai therapy Rats whole blood high cut,low shear viscosity and significantly lower than the model group,and there were no significant change in plasma viscosity(P>0.05);Bai Mai therapy group erythrocyte rigidity index and erythrocyte electrophoresis time,erythrocyte deformation time and erythrocyte aggregation index lower than the model group.They have significant difference(P<0.05),fibrinogen had no significant difference(P>0.05).(4)Inflammation detection:Bai Mai therapy group 1 days and 3 days in the blood IL-1 β,TNF-α is significantly lower than the model group,and have statistical significance(P<0.05),the expression of 3 days less than 1 days;Bai Mai therapy group for 1 days 3 days,the cerebral cortex oflL-1β was significantly lower than that of model group,and 1 days have significant difference(P<0.05);the 1 day,the 3 day Bai Mai therapy group,TNF-a was significantly lower than that of model group,and 3 days was statistically significant(P<0.05).(5)Microvessel regeneration and apoptosis detection:the CD34 content in the Bai Mai therapy group was significantly higher than that in the model group on the 7 day and the 14 day,and there was a significant difference(P<0.05)...
