Depression is a kind of emotional disease with sleep disorders,persistent depression and loss of interest as the main symptoms of the disease,In recent years showed a high incidence,high recurrence rate and high suicide rate characteristics,and gradually showed a trend of younger.According to the latest data in 2017,the total number of depression patients in the world has reached 322 million at present.The overall global prevalence was 4.4%,of which the prevalence of depression in China was 4.2%.The conservative estimate of the number of patients with over 54 million people.However,the recognition rate and treatment rate of depression in China is relatively low,with less than 10%of patients receiving drug treatment.In recent years,with the development of economy and the increase of work and life pressure,the incidence has risen rapidly and the age group has been gradually younger.According to the current data,depression will become the second largest medical problem in the world to be next only to cardiovascular and cerebrovascular diseases by 2030,posing a serious threat to human health.Therefore,depression is a multi-system,multi-mechanism global medical problem.So far,its pathogenesis has not been completely clear.Currently there is a lack of specific methods for the treatment of depression provides the perfect solution.Therapeutic methods are mainly Western medicine and electric shock therapy;traditional Chinese medicine acupuncture therapy acupuncture points through acupuncture and adjust the yin and yang,regulate the role of organs can significantly improve depression patients with anxiety,insomnia and cognitive disorders and other symptoms,and Compared with western medicine antidepressants,acupuncture has multiple targets,multiple levels,no side effects and a series of advantages of high safety.Acupuncture treatment of depression mechanism has been a large number of research results.There has been a great deal of research about the specific mechanism of acupuncture treatment of depression,but the specific therapeutic targets and pathways are not yet clear,so further research and discussion is needed.ObjectiveThis study through the observation and analysis of electricity for chronic stress depression model rats behavior,pathology and ria method to explore the antioxidant effect of electric acupuncture and different brain just cGMP content and the relationship between oxidative stress,to explore the specific mechanism of electric acupuncture treatment of depression mechanism.Methods1.Animal grouping 32 rats were selected and the rats were divided into four groups:normal control group,model group,fluoxetine group,and electroacupuncture group.There were 8 rats in each group,which was adaptable for a week before the experiment.2.Model intervention In addition to the group of rats in the blank group,used the reference method,the rest of the groups were isolated with a single cage and combined with chronic mild unpredictability stress to construct the depression model.The blank group and the rest of the rats were isolated,kept n different rearing room.Rats in each group received 21 days of 7 different stimuli,and 1 was randomly assigned to each day,but the same stimulus did not occur continuously and each stimulus was stimulated at least 3 times.Stimulation methods respectively were 4? cold water swimming 5 min;day and night upside down 24 h;fasting 24 h;humid environment 24h;pinch tail 3 min;no water for 24 h;bound 3 h.3.Treatment methods:Blank group:21 days of normal feeding,normal drinking water intake,do not accept any stimulation;Model group:rats were isolated and accepted 21-day chronic stress modeling treatment;Electroacupuncture group:Electro-acupuncture intervention was performed 1 h before stress.According to "Experimental Acupuncture and Moxibustion Science",flat acupoints Baihui and Yin Tang acupoints were 0.5-1 cm in depth and 1mA in current.,The frequency of electroacupuncture was 2 Hz,the needle was kept for 30 min each time,once a day,other operations were the same as the model group.Fluoxetine group:1 h before the drug intervention,the fluoxetine with physiological saline dubbed 1mg/ml solution,2mg/kg intragastric administration,1 day,other operations with the model group.4.Test indicators and methods.The open field test,weight measurement and sugar consumption test were used to evaluate the behavioral changes of model rats;The effects of electroacupuncture on the pathological changes in the brain regions were evaluated by using the HE staining to observe the pathological changes of the hippocampus and the frontal lobe.The levels of cGMP and SOD and MDA in the hippocampus and the frontal lobe were detected by using radioimmunoassay methods.Results1.Behavioral changThere was no significant difference in behavioral performance between the groups before the experimental stress(p>0.05).After the experimental stress,the body weight,sugar consumption and open field horizontal exercise and vertical exercise blank group were higher than the model group,there was a significant difference(P<0.01);the drug group and the electroacupuncture treatment group rats The above behavioral aspects were higher than those of the model group,with statistical significance(P<0.01).Compared with the drug group,the percentage of sugar water preference in the electroacupuncture group was higher than that of the drug group,which was statistically significant(P<0.05).2.Pathological resultsThe results of HE staining showed that the hippocampal cells in the normal control group had clear structure,regular morphology,large nucleus and round nucleus,clear nucleolus,and dense organization structure;the model group had reduced hippocampal cell layer,unclear structure,and obvious cell degeneration.Irregular in shape,cell shrinking,shrinking death,some nerve cells fall off and forming vacuolization;electroacupuncture group and drug group hippocampal cells are arranged in a regular manner,the cell layer is rich,the polarity is obvious,the cell outline is clear,most nerves The cell membrane is relatively complete.The frontal cells of the normal control rats were arranged in a dense and regular organization with intact neurons and a clear structure.The rats in the model group had reduced levels of frontal cells,unclear structure,obvious cell deformities,loosely arranged cells,and some cells were detached and formed empty.In the electroacupuncture group and the drug group,the frontal lobe cells were arranged more regularly,the morphology of the neurons was better,the neuronal membrane was relatively intact,and the nucleolus was clear.3.Content of MDA,SOD and cGMP in rat hippocampusCompared with the normal control group,the content of MDA and cGMP in the hippocampus of the model group increased significantly(P<0.01),while the expression of MDA and cGMP in the hippocampus of the electroacupuncture group was significantly decreased compared with the model group.SOD increased significantly(P<0.01),and there was no statistical difference compared with blank group(P>0.05).Compared with the normal control group,the expression of SOD in the hippocampus of the model group was significantly reduced(P<0.01),while the relative content of SOD in the hippocampus of the electroacupuncture group rats was significantly higher than that of the model group(P<0.01),statistically significant.4.The content of MDA,SOD and cGMP in the frontal lobe of ratsCompared with the normal control group,the content of MDA and cGMP in the frontal lobe of the model group was significantly increased(P<0.01),with statistical significance(P<0.05);compared with the model group,fluorine The relative expressions of MDA and cGMP in the frontal lobe of the rats of the Cetyl group and the electroacupuncture group were reduced,there was a statistical difference(P<0.05).There was no significant difference in expression between the electroacupuncture group and the fluoxetine group(P>0.05).Compared with the normal control group,the relative content of SOD in the frontal lobe tissue of the model group was significantly reduced(P<0.01);compared with the model group,the frontal lobe tissue of the fluoxetine group and the electroacupuncture group The relative expression content of SOD increased,there was a statistical difference(P<0.05),but there was no significant difference in the expression of electroacupuncture group and fluoxetine group(P>0.05).Conclusion1.Fluoxetine and electro-acupuncture both have anti-depressant effects,and electro-acupuncture can better improve the rat's responsiveness to reward mechanisms,and it has obvious advantages in improving anhedonia.2.Oxidative stress and cGMP and depression there is a significant degree of relevance.3.Electroacupuncture reduces cGMP content in different brain regions of rats may be one of the mechanisms of its antidepressant effect.4.The protective effects of electro-acupuncture on chronic stress-induced depression model rats may be through increasing the body's own antioxidant capacity,regulating the transduction of second messenger cGMP in cells and inhibiting the further damage of cerebral neurons The body's protective effect.
