Based On Rho/Rock Signal Transduction Pathway, The Intervention Study Of Yiqi Xiaozheng Tongluo Recipe On Rat Pulmonary Fibrosis Model Was Studied

Background:Idiopathie pulmonary fibrosis(IPF)is gradually increase after breathing difficulties,irritating cough,asthma suppress as the main clinical manifestations of lesions.The Rho/Rock signaling pathway can regulate cell proliferation,participate in tissue damage repair and regeneration,and play an important role in the process of tissue fibrosis.Inflammation,damage,such as activated Rho,Rho activation signals in Rock activation,Rock-1 content increased,the activation of Rock through a series of phosphory lation process,muscle increased levels of fibroblasts,excessive fibroblast proliferation,transformation,gathered themselves together,and promote the synthesis of various types of inflammatory cells in the synthesis of extracellular matrix.Fibroblasts in the important components of extracellular matrix I,the formation of collagen type Ⅲ imbalance,metabolic disorders play an important role in the process.This research group combined with the theory of "jusan jingluo zhengjia "of traditional Chinese medicine,first proposed the"Micro Zheng jia in lung collateral" pathogenesis hypothesis of COPD,based on the pathogenesis hypothesis,formulated the therapeutic principle yiqixiaozheng huoxuetongluo,and drew up yiqi xiaozheng tongluo formula.Satisfactory results were obtained in clinical and early animal experiments.Based on many years of clinical observation,the teacher found that IPF had a common pathological basis with chronic obstructive pulmonary disease(COPD),and it was also effective in treating IPF patients.Research methods:Experiment using disposable endotracheal intubation BLM made mold injection preparation of pulmonary fibrosis in rats model of interventional treatment in yiqi xiaozheng tongluo formula,prednisone as the positive control drug,observe the mental state of rats during experiment and nose and mouth secretions,weight,feeding water quantity,fur color,such as respiratory general situation,within 28 days after the building to be put to death animals,separation of lung tissue,observe the shape change,preparation of pathological,color using HE staining and Masson trichromatic dye,then view pathology change,using the ELISA method to observe the serum Ⅰ,Ⅲ collagen content,Rock-1,RT-PCR detection of lung tissue in Rock-1mRNA transcription level.Results:Yiqi xiaozheng tongluo formula each dosage group of rats in mental state,the response sensitivity,fur onyx glossiness,did not see nose and mouth secretions,breathing rhythms,feeding,the performance of many aspects,such as drinking water,the average weight is better than the model group.The structural integrity of bronchial and alveolar cavity,inflammatory cell infiltration,fibroblast aggregation,and collagen deposition were all less than that of the model group.Test results:the serum Ⅰ,Ⅲ collagen,Rock and transcriptional level of rock-1 mRNA in lung tissues transcription yiqi xiaozheng tongluo formula reduce each dose group compared with model group,P<0.05,statistically significant differences.Research conclusion:Yiqi xiaozheng tongluo formula can lighten the BLM general symptoms of model rats,decrease the Rho/Rock Rock-1 mRNA expression in the channel,reduce the BLM model rats Ⅰ,Ⅲ type collagen formation,reduce the degree of pulmonary fibrosis in rats.Therefore,we believe that tongluo can prevent and treat pulmonary fibrosis.
