Part1: Polymorphism of Sp110 and TLR-2 gene and susceptibility to tuberculosis in Tibetan populationObjective Study distribution of single nucleotide rs1135791,rs72255,rs9061(Sp110 gene)polymorphism and rs5743708(TLR-2 gene)polymorphism in Tibetan population,and exam correlation between 4 single nucleotides’ polymorphism and tuberculosis prevalence of native Tibetan population,in order to provide genetic evidence for predicting tuberculosis risk of the Tibetan population.Method 1.A case-control study was carried out in the department of pulmonary medicine,the Third People’s Hospital,Tibet autonomous region.343 tuberculosis patients(an average age of 31.09±16.23)were diagnosed by positive tuberculosis sputum smear.In the case group,187 patients were male and 156 patients were female.The control group was consist by 260 healthy Tibetan individuals(an average age of 32±16.39),who were matched with case group by age and gender.In the control group,137 individuals were male and 123 individuals were female.2.i MLDR was adopted to detect polymorphism of single nucleotide rs1135791,rs722555,rs9061(Sp110 gene)and genotype frequency and allele frequency distribution of single nucleotide rs5743708(TLR-2 gene).3.Statistical analysis method: using Excel 2007 and SPSS20.0 statistical software for data entry and analysis,the case group and the control group were compared with chi square test(x2)and t,We use Chi-square test and multivariate regression analysis to exam correlation between single nucleotide polymorphism of Sp110 gene and TLR-2 gene and susceptibility to pulmonary tuberculosis of Tibetan population.Result 1.Frequency of T allele(single nucleotide polymorphism rs1135791,Sp110 gene)in the case group and the control group were 78.1%and 79.8% meanwhile,the frequency of C allele in the case group and the control group were 21.9% and 20.2%.There was no significant difference of allele distribution between the two groups(x2=0.50,p=0.48).In the case group,frequencies of T/T,T/C and C/C genotype(single nucleotide rs1135791)were 61.5%,33.2%,5.3%;in the control group were 63.5%,32.7%,3.8%.Genotype distribution between the two groups showed no significant difference(x2=0.73;p=0.69).Frequencies of G allele(single nucleotide rs722555,Sp110 gene)in the case group and the control group were respectively 57.4% and 57.5%,the frenquencies of A allele in the case group and the control group in the 42.6% and 42.5%.There was no significant difference of allele distribution between the two groups(x2=0.0005;p=0.98).Frequencies of G/G,G/A and A/A genotype(aingle nucleotide rs722555)in the case group were 36.1%,42.6%,21.3%;and in the control group were 32.7%,49.6%,17.7%.Genotype distribution between the two groups showed no significant difference(x2=3.09;p=0.21).Frequencies of G allele(single nucleotide rs9061,Sp110 gene)in the case group and the control group were respectively 91.7% and 89.6%,while the frequencies of A allele in the case group and the control group were 8.3% and10.4%.There was no significant difference of allele distribution between the two groups(x2=1.53;p=0.22).Frequencies of G/G,G/A and A/A genotype(single nucleotide rs9061)in the case group were 83.4%,16.6%,0%;and in the control group were 80.8%,17.7%,1.5%.Genotype distribution between the two groups showed no significant difference(x2=3.09;p=0.21).The distribution of single nucleotide rs9061 was statistically different under the recessive genetic model(OR=6.022;p=0.021).2.Only GG genotype(single nucleotide rs5743708,TLR-2 gene)was detected in the case group and the control group.There was not G/A or A/A.The result indicated that the single nucleotide rs5743708 of TLR-2 gene was not polymorphic in Tibetan population.Conclusion : Distributions of single nucleotide rs1135791,rs72255,rs9061(Sp110 gene)polymorphism and rs5743708(TLR-2 gene)polymorphism were not associated with tuberculosis prevalence of native Tibetan population.Polymorphism of the 4 single nucleotides may not be a risk factor for tuberculosis in Tibetan population.Part2:Investigation on knowledge and attitude of tuberculosisprevention and control among senior high school studentsin LhasaObjective To understand the knowledge and attitude of tuberculosis in senior high school students in Lhasa and the main ways to acquire TB knowledge,in order to better carry out the school tuberculosis health education provide reference.Methods A total of 729 students from 6 high schools in Lhasa were investigated by stratified random cluster sampling to investigate the knowledge,attitude and the source of tuberculosis prevention and control knowledge.Using SPSS 20 statistical software for data entry and analysis,in which the measurement data were compared using t test and F test,descriptive analysis using x2 test.Results The overall awareness rate of tuberculosis prevention and control knowledge was 45.6%,the awareness rate of tuberculosis was 1.5%,the attitude of tuberculosis was 60.3%,Han students’ knowledge score(t=7.145,p = 0.001),attitude score(t=5.179,p = 0.001)were higher than those of Tibetan students;The liberal arts students’ knowledge score(t =3.471,p = 0.001)higher than the science students;urban students’ knowledge score of TB(F=26.841,p < 0.001),attitude scores(F=19.160,p=0.001)were higher than that of the students in agricultural and pastoral areas;The main source of knowledge of TB prevention and control is "TV",the most popular anti-TB promotional material is "picture-based promotional materials".Conclusion The awareness rate and attitudes of tuberculosis prevention and control knowledge among the high school students in Lhasa,Tibet are all low,so it is necessary to popularize the health education of tuberculosis prevention and control knowledge for school-oriented high school students.
